摘  要:探究鹿心肽对小鼠酒精性肝肾损伤的影响。选用20只雄性健康昆明种小鼠,随机分为空白组、对照组、鹿心肽低剂量、高剂量组4组。除空白组外,其他3组灌胃56度白酒,建立酒精性肝肾损伤模型。称量小鼠体重及器官湿重,测得肝肾指数,通过检测血清中抗氧化指标丙二醛(MDA)、超氧化物歧化酶(SOD)含量以检测氧化应激水平,并使用酶标仪及试剂盒对乙醛脱氢酶(ALDH)活性进行检测;在用Trizol取得肝肾总RNA后用qPCR检测肝肾脏白介素-1β (IL-1β)、白介素-6 (IL-6)及肿瘤坏死因子(TNF-α) mRNA表达水平。结果表明:与对照组对比,鹿心肽高低剂量组在肝脏指数上均有显著下降,鹿心肽高低剂量的肝脏指数也显著低于对照组,而低剂量肝脏指数明显低于高剂量组;经检测显示,空白组与对照组的肾脏指数有极显著差异,而对照组与低剂量组有极显著差异,与高剂量组则为显著差异;在SOD含量方面,鹿心肽高低剂量组相对对照组均有不同程度上升;相比对照组,鹿心肽高低剂量组MDA含量均有降低;与对照组相比,鹿心肽高低剂量组在ALDH数值上均有极显著上升;在TNF-α、IL-1β、IL-6的表达水平上,鹿心肽高低剂量组相对于对照组有显著下降。说明鹿心肽对小鼠酒精性肝肾损伤有一定的干预与预防作用。Explore the effect of deer heart peptide on alcoholic liver and kidney injury in mice.Twenty healthy male Kunming mice were randomly divided into blank group,control group,low dose and high dose groups.Except the blank group,the other 3 groups were administrated with 56-degree liquor to establish the alcoholic liver and kidney injury model.The body weight and organ wet weight of mice were weighed,the liver and kidney indexes were measured,the levels of oxidative stress were detected by detecting the contents of serum antioxidant indexes malondialdehyde(MDA) and superoxide dismutase(SOD),and the activity of acetaldehyde dehydrogenase(ALDH) was detected by enzyme label and kit.The mRNA expression levels of interleukin-1β(IL-1β),interleukin-6(IL-6) and tumor necrosis factor(TNF-α) were detected by qPCR after the total RNA of liver and kidney was obtained by Trizol.The results showed as follows:Compared with the control group,the liver index of high and low dose of deer heart peptide group decreased significantly,the liver index of high and low dose of deer heart peptide group was significantly lower than that of the control group,and the liver index of low dose was significantly lower than that of high dose group.The results showed that the kidney index of the blank group was significantly different from that of the control group,while that of the control group was significantly different from that of the low-dose group and that of the high-dose group.The SOD content in the high and low dose groups was higher than that in the control group.Compared with the control group,MDA content in high and low dose groups was decreased.Compared with the control group,ALDH values in high and low dose groups were significantly increased.The expression levels of TNF-α,IL-1β and IL-6 in the high-low dose group were significantly decreased,compared with the control group.These results indicated that deer heart peptide could intervene and prevent alcoholic liver and kidney injury in mice.
