摘  要:目的探讨蛇床子素(OST)对4T1细胞放疗增敏的作用及其机制。方法4T1细胞经不同浓度OST(25,50,75,100,125,150,175,200μmol/L)处理24 h后用CCK-8法检测细胞活力。将4T1细胞分为IR组和OST(50μmol/L)+IR组,采用克隆平板形成实验检测放疗增敏影响。将4T1细胞分为对照组、OST组(50μmol/L)、IR组(2 Gy)、OST(50μmol/L)+IR(2 Gy)组,TUNEL染色检测细胞凋亡,Western blot检测细胞内Nrf2及NQO1蛋白表达水平变化。结果与0μmol/L OST组相比,不同浓度OST(25,50,75,100,125,150,175,200μmol/L)组4T1细胞的增殖活力明显降低(P<0.05)。与IR组相比,OST+IR组细胞克隆存活分数显著降低(P<0.05);与IR组相比,OST+IR组细胞的增敏比为1.14。与对照组相比,OST组和IR组细胞凋亡率均增加(P<0.05);与IR组相比,OST+IR组细胞凋亡率明显增加(P<0.05)。与对照组相比,OST组Nrf2及NQO1蛋白表达水平明显增加(P<0.05),IR组Nrf2及NQO1蛋白表达水平明显降低(P<0.05);与IR组相比,OST+IR组Nrf2及NQO1蛋白表达水平明显降低(P<0.05)。结论蛇床子素可以增加乳腺癌细胞的放疗敏感性,其可能与Nrf2/NQO1通路机制有关。Objective To investigate the effect and mechanism of osthol(OST)on radiosensitization of 4T1 cells.Methods 4T1 cells were treated with different concentrations of OST(25,50,75,100,125,150,175,200μmol/L)for 24 h,and then the cell viability was detected by CCK-8.4T1 cells were divided into IR group and OST(50μmol/L)+IR group,and then clone plate formation assay was used to detect the effect of radiotherapy sensitization.4T1 cells were divided into control group,OST(50μmol/L)group,IR(2 Gy)group,OST(50μmol/L)+IR(2 Gy)group.TUNEL staining was used to detect the apoptosis,and Western blot was used to detect Nrf2 and NQO1 protein expression levels in 4T1 cells.Results Compared with 0μmol/L OST group,the prolife-ration activity of 4T1 cells was significantly decreased in different OST groups(25,50,75,100,125,150,175,200μmol/L)(P<0.05).Compared with IR group,the cell clonal survival fraction in OST+IR group was significantly decreased(P<0.05).Compared with IR group,the sensitization ratio of cells in OST+IR group was 1.14.Compared with control group,the apoptosis rate was increased in OST group and IR group(P<0.05).Compared with IR group,the apoptosis rate was significantly increased in OST+IR group(P<0.05).Compared with control group,the protein expression levels of Nrf2 and NQO1 were significantly increased in OST group(P<0.05),while the protein expression levels of Nrf2 and NQO1 was significantly decreased in IR group(P<0.05).Compared with IR group,the expressions of Nrf2 and NQO1 were significantly decreased in OST+IR group(P<0.05).Conclusion Osthol can increase the radiosensitivity of breast cancer cells via the Nrf2/NQO1 pathway.
