摘  要:非酒精性脂肪性肝病(non-alcoholic fatty liver disease,NAFLD)是指排除酒精及其他损伤肝脏的因素外所导致的以肝脏脂肪沉积为特征的与代谢紊乱密切相关的临床综合征,包括单纯性脂肪肝(simple steatohepatitis,SFL)、非酒精性脂肪性肝炎(non-alcoholic steatohepatitis,NASH)和由此发展而来的肝硬化。流行病学统计提示我国NAFLD患病率逐年上升,已成为我国最常见的慢性肝脏疾病。部分NAFLD患者可进展为NASH,NASH又可进一步发展为肝细胞癌(hepatocellular carcinoma,HCC),是导致HCC的主要原因,我国人口基数大,由此产生的医疗及经济负担不容小觑。而关于NAFLD的研究离不开模型的构建,更为优良的模型意味着更可靠的研究结果,然而NAFLD造模方法始终无统一的模式。本文回顾了现存的细胞及动物NAFLD模型(涉及模型的构建方式),对各种饮食方式诱导NAFLD动物模型进行了对比,旨在为广大研究人员选择并构造合适的NAFLD模型提供指导。Non-alcoholic fatty liver disease(NAFLD)is a clinical syndrome characterized by fatty deposition of the liver and closely associated with metabolic disorders,excluding alcohol and other liver damaging factors,including simple steatohepatitis(SFL),non-alcoholic steatohepatitis(NASH),and subsequent cirrhosis.Epidemiological statistics indicate that the prevalence of NAFLD is increasing year by year in China,and NAFLD has become the most common chronic liver disease in China.Some NAFLD patients can progress to NASH,and NASH can further develop into hepatocellular carcinoma(HCC),which is the main cause of HCC.China has a large population base,and the resulting medical and economic burden should not be underestimated.Research on NAFLD is inseparable from the construction of models.A better model means more reliable research results.However,there is no unified model for modeling NAFLD.In this paper,existing cell and animal NAFLD models were reviewed,including model construction methods,and animal NAFLD models induced by various dietary patterns were compared,aiming to provide guidance for researchers to select and construct appropriate NAFLD models.
