摘  要:目的通过电感耦合等离子体-质谱(ICP-MS)法建立同时测定秀丽隐杆线虫体内26种元素含量的方法,同时分析线虫生长发育不同阶段体内元素的含量变化。方法以65%~68%HNO3作为消解液,采用湿法消解处理线虫,利用ICP-MS同时测定线虫中26种元素Al、As、B、Ba、Bi、Ca、Cd、Co、Cr、Cu、Fe、Ga、In、K、Mg、Mn、Mo、Na、Ni、Pb、Sc、Se、Te、V、Zn、Zr。将线虫同步化后获得的卵均匀分成5组,分别培养至L1期、L2期、L3期、L4期以及Adult期,应用建立的ICP-MS法测定26种元素含量。结果26种元素在0~500μg·L^(-1)线性关系良好,R2≥0.9993;仪器检出限为0.0001~0.0903μg·L^(-1);仪器精密度(RSD)为0.4%~1.8%;重复性(RSD)为0.0%~2.5%;稳定性(RSD)为0.8%~4.6%;平均加标回收率为80.8%~118.5%,RSD为0.2%~1.5%。L4期线虫中总体元素含量最高,其次是L2期,L1期线虫中总体元素含量最低。结论通过ICP-MS测定了线虫生长发育过程中体内元素的含量,该方法快捷灵敏、简便高效,适用于对线虫中元素的分布情况进行分析,为进一步研究单一元素或多种元素对秀丽隐杆线虫的潜在影响提供参考。Objective A method was established to simultaneously determine the content of 26 elements in C.elegans by Inductively Coupled Plasma Mass Spectrometry(ICP-MS),and to analyze the content changes of elements in different stages of C.elegans growth and development.Methods The nematodes were treated by wet digestion using 65%—68%HNO3 as the digestion solution,and 26 elements(Al,As,B,Ba,Bi,Ca,Cd,Co,Cr,Cu,Fe,Ga,In,K,Mg,Mn,Mo,Na,Ni,Pb,Sc,Se,Te,V,Zn,and Zr)in the nematodes were determined simultaneously using ICP-MS.The eggs obtained after synchronization of nematodes were evenly divided into five groups and cultured to L1,L2,L3,L4,and Adult stages,respectively.The content of 26 elements was determined using the established ICP-MS method.Results The results showed that the linearity of the 26 elements was good in the range of 0—500μg·L^(-1) with R2≥0.9993,the instrumental limits of detection were 0.0001—0.0903μg·L^(-1),the instrumental precision RSD was 0.4%—1.8%,the reproducibility RSD was 0.0%—2.5%,the stability RSD was 0.8%—4.6%,the average spiked recoveries were 80.8%—118.5%,and the RSD was 0.2%—1.5%.The L4 stage nematodes have the highest overall element content,followed by the L2 stage,with the L1 stage nematodes having the lowest overall element content.Conclusion The content of elements in nematodes during growth and development was determined by ICP-MS,which is a fast,sensitive,simple and efficient method suitable for analyzing the distribution of elements in nematodes,and provides a certain theoretical basis for the further study of the potential effects of a single element or a variety of elements on Caenorhabditis elegans.
