摘 要:研究桔梗总皂苷对痛风性关节炎小鼠的影响及基于NLRP3/ASC/caspase-1途径的抗炎作用。将小鼠分为空白组、模型组、秋水仙碱(1 mg/kg)组和桔梗皂苷(25、50、100 mg/kg)组,连续灌胃7 d。在第5 d灌胃1 h后,将小鼠麻醉,踝关节处消毒后将MSU注射到关节中。通过测量关节肿胀程度和检测炎症因子、过氧化物因子和相关蛋白的表达水平,阐明桔梗总皂苷治疗痛风性关节炎的抗炎作用。结果表明,桔梗总皂苷能有效减轻肿胀,降低炎症因子和过氧化物水平。桔梗总皂苷能够改变炎症区域的细胞形态,踝关节处的NLRP3、ASC、caspase-1和IL-1β的蛋白质水平也显著降低。因此桔梗总皂苷能有效缓解MSU引起的痛风性关节炎,其机制与NLRP3/ASC/caspase-1途径上的蛋白减少有关。To study the effects of total saponin of Platycodon grandiflorum on mice with gouty arthritis and its antiinflammatory effects based on NLRP3/ASC/caspase-1 pathway.Mice were divided into blank group,model group,colchicine(1 mg/kg)group and orris saponin(25,50,100 mg/kg)group by gavage for 7 d.After gavage for 1 h on the 5th day,mice were anesthetized and MSU was injected into mice joints after sterilization at the ankle joint.The anti-inflammatory effects of total saponin from Platycodon grandiflorum in the treatment of gouty arthritis were elucidated by measuring the degree of joint swelling and detecting the expression levels of inflammatory factors,peroxide factors and related proteins.The results showed that total saponin was effective in reducing the swelling and decreasing the levels of inflammatory factors and peroxides in mice,and the pathological sections showed that Platycodon grandiflorus total saponin was able to change the cell morphology of the inflammatory area.The protein level of NLRP3,ASC,caspase-1,and IL-1βat the ankle joint decreased too.The results indicated that Platycodon grandiflorum total saponin could effectively alleviate MSU-induced gouty arthritis by a mechanism related to the reduction of proteins in the NLRP3/ASC/caspase-1 pathway.
