摘 要:以获取对松树天牛高致病的生防真菌为目标,从黑龙江省森林土壤中初步分离得到18株虫生真菌,分别采用爬行法测定其对松树天牛幼虫、成虫的致病力,同时测定高致病力菌株的菌丝生长速度和产孢量,对高致病力菌株进行种类鉴定。结果表明:18株虫生真菌对松树天牛幼虫、成虫均表现出不同程度的致病力。其中菌株M14-1、H_(2)表现出较强的致病力,在处理天牛幼虫、成虫的6 d,累计死亡率均达到了100%,菌株M14-1、H_(2)菌落生长速度快,产孢量大;在90 mm×15 mm(直径×深度)的马铃薯葡萄糖琼脂(PDA)培养基的培养皿上培养15 d时,菌落直径分别为5.68、4.91 cm,产孢量分别为3.86×10^(8)、4.1×10^(8)个·板^(-1)。在孢子浓度为10^(7)个·mL^(-1)时,菌株M14-1、H_(2)对松树天牛成虫的半致死时间(LT_(50))分别为4.15、4.31 d。经形态学和分子生物学鉴定,菌株M14-1、H_(2)皆属于金龟子绿僵菌(Mertarhizium anisopliae)。The goal of this study was to obtain entomopathogenic fungi with high pathogenicity against the pine long-horned beetle from forest soil in Heilongjiang Province. Eighteen strains of entomopathogenic fungi were isolated and their pathogenicity against pine long-horned beetle larvae and adults was determined using a crawling assay. The strains with high pathogenicity were also evaluated for their mycelial growth rate and spore production, and identified at the species level. The results showed that all 18 strains of entomopathogenic fungi exhibited varying degrees of pathogenicity against pine long-horned beetle larvae and adults. Among them, strains M14-1 and H2 showed strong pathogenicity, with a cumulative mortality rate of 100% for both larvae and adults after 6 days of treatment. Strains M14-1 and H2 had fast colony growth and large spore production. When cultured on PDA medium of 90 mm×15 mm (diameter×depth) for 15 days, the colony diameters were 5.68 cm and 4.91 cm, respectively, and sporulation yields were 3.86×10^(8) and 4.1×10^(8) per plate, respectively. The half lethal time (LT _(50) ) of strains M14-1 and H_(2) against pine long-horned beetle adults was 4.15 and 4.31 days, respectively, at a spore concentration of 10^(7) spores/mL. Morphological and molecular biology identification confirmed that strains M14-1 and H2 belong to Metarhizium anisopliae .
