酶法提取蛋壳膜中胶原蛋白的工艺优化

摘　　要：胶原蛋白具有促进伤口愈合、止血等功能。目前市场上胶原蛋白的主要来源是阿胶及各种动物的皮胶等。用蛋壳膜也可提取胶原蛋白,但相关研究较少。本研究以废弃蛋壳为原材料,先使用盐酸浸泡使壳膜分离,用氢氧化钠将蛋白质进行纯化,采用胃蛋白酶酶解-盐析法提取废弃蛋壳膜中的胶原蛋白。以胃蛋白酶的浓度、溶液pH值、酶解时间、提取温度四个因素为变量进行单因素实验,在此基础上,通过L9(34)正交实验设计得出最佳的提取工艺条件,用紫外分光光度法测定蛋壳膜中胶原蛋白的含量。研究结果表明:酶用量10%、溶液p H值1.7、酶解时间30 h、提取温度40℃,胶原蛋白的提取率是30.41%。相较有关研究,提取率提高了0.361%。Collagen has the functions of promoting wound healing and hemostasis.At present,the sources of collagen on the market are done-hide gelatin and various animal skin adhesives.Collagen can also be extracted from eggshell membrane,but there are few related studies.The waste eggshell was used as raw material,firstly soaked in hydrochloric acid,the shell membrane was separated,the protein was purified with sodium hydroxide,and the collagen in the waste eggshell membrane was extracted by pepsin enzymolysis and salting out method.The concentration of pepsin,pH,time and temperature were investigated for single factor test,and then the optimal extraction process was obtained by L9(34)orthogonal test.The content of collagen in eggshell film was determined by UV spectrophotometry.The results showed that the extraction rate of collagen from egg shell membrane was 30.41%under the conditions of 40℃,pH1.7,30 h enzymolysis reaction time and 10%enzyme dosage.Compared with related studies,the extraction rate increased by 0.361%.
