摘 要:建立一种婴幼儿乳粉样品的简便快速处理方法,以高效液相色谱法定量检测样品中5种糖含量。样品经水充分溶解,沉淀蛋白质后,注入高效液相色谱仪进行检测。果糖、葡萄糖、蔗糖、麦芽糖和乳糖的质量浓度在1~10 mg/mL范围内线性良好,线性相关系数均大于0.999,方法检出限均为0.15%,定量限均为0.45%。在1%、2%、10%的加标水平下,果糖、葡萄糖、蔗糖、麦芽糖、乳糖测定结果的相对标准偏差为0.9%~2.6%(n=6),回收率分别为96.5%~99.4%、97.0%~101.0%、98.2%~100.8%、99.6%~101.5%、95.5%~99.6%。该方法可用于婴幼儿乳粉中5种糖含量的日常检测。A simple and rapid pre-treatment method for infant formula milk powder samples was established,and a high performance liquid chromatography(HPLC)method was developed for the quantitative determination of 5 sugars in the samples.The samples were dissolved in water,the proteins were precipitated,and then injected into high performance liquid chromatography for detection.Fructose,glucose,sucrose,maltose and lactose showed good linearity in the range of 1-10 mg/mL with the correlation coefficients were more than 0.999.The detection limits were all 0.15%,and the quantitation limits were all 0.45%.The recoveries of fructose,glucose,sucrose,maltose,and lactose were 96.5%-99.4%,97.0%-101.0%,98.2%-100.8%,99.6%-101.5%,and 95.5%-99.6%at the addition level of 1%,2%and 10%,respectively,with the relative standard deviations of 0.9%-2.6%(n=6).The method can be used for the daily determination of 5 sugars in infant formula milk powder.
