摘 要:多色免疫组织化学(Multiplex immunohistochemistry,mIHC)技术是一种新型多靶点病理组织染色、成像及分析技术。该技术通过在单张组织切片上检测多种生物标志物的表达水平及空间分布,实现对细胞的表型、组成、形态及细胞间相互作用机理的全面解析。近年来,mIHC技术被成功应用于肿瘤免疫研究领域,为肿瘤的诊断和治疗开辟了新的途径。本文从近年开发的mIHC检测技术出发,针对不同类型mIHC技术的检测原理及其特点进行了详细阐述,重点讨论了新型mIHC检测技术在肿瘤标志物及肿瘤微环境检测等领域中的应用,并对mIHC在肿瘤免疫诊断和治疗领域的应用前景和发展趋势进行了展望。Multiplex immunohistochemistry is a novel pathological tissue staining imaging and analysis technology.It can comprehensively analyze the cellular phenotype,composition and morphology by simultaneously analyzing biomarkers'description and spatial distribution on a single tissue section.In the last ten years,this technology has been successfully applied in tumor immunity research,which opens a new way to diagnose tumors.In this review,based on the recently developed multiplex immunohistochemical technology,several new techniques for detecting tumor markers for immunodiagnostic purposes and analyzing the tumor microenvironment were reviewed.The research prospects in multiplex immunohistochemistry in tumor diagnosis and therapy were also discussed.
