摘  要:目的探究五味子酚(Schisanhenol)对肝细胞癌细胞增殖及作用机制的影响。方法将HepG2细胞分为空白对照组和五味子酚3、10、30μmol/L 3个浓度组。应用Western Blot检测HepG2细胞PD-L1蛋白表达;通过免疫荧光试验进一步检测五味子酚对PD-L1蛋白表达的影响;通过集落试验观察HepG2细胞的增殖情况。结果HepG2细胞PD-L1蛋白表达结果显示,3、10、30μmol/L五味子酚均可显著抑制PD-L1蛋白表达(P<0.05或P<0.01),且抑制效果呈剂量依赖性;免疫荧光试验和集落试验结果显示,30μmol/L五味子酚对HepG2细胞增殖有显著的抑制作用(P<0.01)。结论五味子酚通过调节PD-L1蛋白表达抑制肝细胞癌细胞HepG2增殖。To investigate the effect of Schisanhenol on the proliferation of hepatocellular carcinoma cells and its related mechanism.[WTHZ]Method[WT][KG1]Hepatocellular carcinoma HepG2 cells were divided into blank control group and three Schisanhenol groups(3μmol/L,10μmol/L and 30μmol/L).The expressions of PD-L1 proteins in HepG2 cells were detected by Western Blot.Immunofluorescence assay was used to further detect the effect of Schisanhenol on PD-L1 protein.The proliferation of HepG2 cells were observed by cell colony assay.[WTHZ]Results[WT][KG1]The expression results of PD-L1 proteins in HepG2 cells showed that Schisanhenol with 3,10 and 30μmol/L could inhibit the expression of PD-L1 proteins(P<0.05 or P<0.01);The results of immunofluorescence assay and colony assay showed that Schisanhenol with 30μmol/L could obviously inhibit the proliferation of HepG2 cells(P<0.01).[WTHZ]Conclusion[WT][KG1]Schisanhenol inhibits the proliferation of liver cancer cell HepG2 through regulating PD-L1 protein.
