摘 要:目的探讨信号肽酶复合体亚基2(SPCS 2)基因在乳头状甲状腺癌(papillary thyroid carcinoma,PTC)组织中的表达水平及其意义。方法利用UALCAN及HPA数据分析平台评估PTC组织和正常组织中SPCS 2基因表达水平。在Gepia数据库预测SPCS 2基因表达水平对PTC患者无病生存时间的影响。Cox回归模型分析影响PTC患者预后的独立危险因素。并对该基因进行KEGG、GO和GSEA分析,确定参与PTC的重要信号通路。结果PTC患者肿瘤组织中SPCS 2基因表达水平均显著低于正常组织(Z=-43.219,P<0.001)。与SPCS 2低表达的PTC患者相比,SPCS 2基因高表达组PTC患者无病生存时间更长(P<0.05)。Cox回归模型分析显示,PTC患者肿瘤组织中SPCS 2低表达是导致PTC患者预后不良的独立危险因素(HR=0.573,P<0.05)。富集分析预测显示,线粒体氧化磷酸化通路富集在SPCS2高表达组。结论PTC患者肿瘤组织中SPCS 2基因表达水平下调,SPCS 2基因可能参与线粒体氧化磷酸化信号通路的调控,该基因可能是评估PTC患者预后的重要生物标志物。Objective To investigate the expression level and significance of the signal peptidase complex subunit 2(SPCS 2)gene in papillary thyroid carcinoma(PTC).Methods UALCAN and HPA Databases were used to investigate the mRNA expression levels of SPCS 2 in PTC tissue and normal tissue.Gepia Database was used to predict the influence of the expression level of SPCS 2 on the disease-free survival of PTC patients.The Cox regression model was used to investigate the independent risk factors for the prognosis of PTC patients.GO,KEGG,and GSEA analyses were performed for SPCS 2 to identify the important signaling pathways involved in PTC.Results The mRNA expression levels of SPCS 2 in PTC tissue were significantly lo-wer than those in normal tissue(Z=-43.219,P<0.001).The PTC patients with a high expression level of SPCS 2 had a longer disease-free survival time than those with a low expression level of SPCS 2(P<0.05).The Cox regression analysis showed that the low expression level SPCS 2 in PTC patients was an independent risk factor for the poor prognosis of PTC patients(HR=0.573,P<0.05).The enrichment analysis showed that the oxidative phosphorylation pathway of mitochondria was enriched in the SPCS 2 high expression group.Conclusion The expression level of SPCS 2 is downregulated in PTC patients,and SPCS 2 may participate in the regulation of the oxidative phosphorylation pathway of mitochondria.SPCS 2 may be an important biomarker for evaluating the prognosis of PTC patients.
