摘 要:选用50只35日龄家兔随机分为5组,每组10只。对照组(A组)和模型组(B组)正常饲喂,另外3组(C、D、E组)分别在日粮中添加0.2、0.4、0.6mg/kg硒代蛋氨酸,饲养第17天起A组灌喂橄榄油,B组灌喂0.3mg/kg黄曲霉毒素B1(AFB1),C、D、E组正常饲喂含硒代蛋氨酸日粮同时灌喂0.3mg/kg的AFB1,连续攻毒5d后解剖兔取其胃组织,采用HE染色、PAS染色和抗氧化试剂盒研究胃组织形态学变化及其总抗氧化能力(total antioxidant capacity,T-AOC)、总超氧化物岐化酶(total superoxide dismutase,T-SOD)、丙二醛(MDA)的变化特点。HE染色结果显示,与A组相比,其余各组胃黏膜层明显受损,导致胃黏膜层变薄、胃小凹深度降低,尤以E组深度显著降低;PAS染色结果显示,与A组相比,B组阳性层厚度减少、分布不均匀,C、D组胃部PAS阳性层厚度增加,E组阳性层厚度与B组相比变化不明显;抗氧化指标检测结果显示,与A组相比,B组MDA显著升高(P<0.05),C组与B组相比TAOC、T-SOD显著升高(P<0.05),与A组相比,E组T-SOD显著降低(P<0.05)。结果表明,适量有机硒对AFB1致家兔胃黏膜损伤具有一定的保护作用。The purpose of this study is to investigate whether organic selenium can protect the gastric mucosa of rabbits from AFB1injury.Fifty 35-day-old rabbits were randomly divided into 5 groups with 10rabbits in each group.The control group(group A)and the model group(group B)were fed normally,and the other three groups(groups C,D,and E)were fed with 0.2,0.4,and 0.6 mg/kg selenomethionine,respectively.From the 17th day of feeding,group A was fed olive oil,and group B was fed 0.3mg/kg AFB1.Groups C,D,and E were fed with diet containing selenomethionine and 0.3mg/kg AFB1.After 5days of continuous poisoning,the gastric tissue of rabbits was collected,and HE staining,PAS staining and antioxidant kits were used to study the morphological changes of gastric tissue and the characteristics of changes in T-AOC,T-SOD and MDA.The HE staining results showed that compared with group A,the gastric mucosa of the other groups was significantly damaged,leading to the thinning of the gastric mucosa and the reduction of the depth of gastric pits,especially in group E.The PAS staining results showed that,compared with group A,the thickness of the positive layer in group B decreased and was unevenly distributed,the thickness of PAS positive layer in the stomach of groups C and D increased,and the thickness of positive layer in group E did not show significant changes compared to group B.Antioxidant indexes test results showed that compared with group A,group B had a significant increase in MDA(P<0.05),and group C had significant increase in T-AOC and SOD compared with group B(P<0.05),group E had a significant decrease in T-SOD(P<0.05).The results of this experiment confirmed that an appropriate amount of organic selenium had a certain protective effect on AFB1-induced gastric mucosal injury in rabbits.
