摘 要:对栀子黄色素的纯化工艺条件进行优化。以闽东福鼎的栀子为原料,通过大孔树脂的筛选,研究洗脱剂浓度和流速对吸附过程的影响,确定大孔树脂纯化栀子黄色素的新工艺。结果表明,HPD-400型大孔树脂对栀子黄色素的吸附量大,解析率高,纯化效果较好;最佳柱层析条件为洗脱剂质量分数80%,洗脱流速1.0 mL/min。将洗脱液浓缩,真空干燥即得高纯度栀子黄色素,所得黄色素OD_(238 nm/440 nm)比值由1.86降到0.27,色价达到510。为纯化栀子黄色素提供了一种好方法。To optimize the purification process of gardenia yellow pigment.The gardenia of Fuding in eastern Fujian was used as raw material,and a new method for purification of gardenia yellow pigment with macroporous resin was determined.The effect of concentration and flow rate of eluent on the adsorption process was studied through resin screening.The results showed that the macroporous resin HPD-400 had large adsorption capacity,high resolution and good purification effect for gardenia yellow pigment.The optimum column chromatographic conditions were 80%concentration of eluent and 1.0 mL/min elution flow rate.The high purity gardenia yellow pigment was obtained by concentrating the eluent and vacuum drying.The ratio of OD_(238 nm/440 nm)of the yellow pigment was reduced from 1.86 to 0.27,and the color value was 510.This paper developed a good method for purification of gardenia yellow pigment.
