摘  要:目的 筛选胰腺癌中的核心甲基化驱动基因,探究其生物功能及临床意义。方法 基于TCGA/GEO数据库中胰腺癌的mRNA-seq数据及匹配的临床数据,利用R分析软件相关数据包及部分在线分析工具,分析并获得胰腺癌中甲基化驱动基因;采用Lasso-Cox回归分析筛选核心甲基化驱动基因,并探究其在胰腺癌中的功能特点及临床意义;最后基于核心甲基化驱动基因构建预后模型并进行相关探究。结果 甲基化驱动基因ZSCAN18、FERMT1、LIPH、LAMA3及S100A16对胰腺癌表现出良好的诊断效能,且FERMT1、LIPH、LAMA3及S100A16的表达上调与患者较差的预后相关(P<0.05),ZSCAN18的表达上调则与患者较好的生存预后相关(P<0.05),而基于甲基化驱动基因ZSCAN18、FERMT1、LIPH、LAMA3及S100A16的预后模型表现出良好的预测效能,具有较好的临床应用潜能。结论 甲基化驱动基因ZSCAN18、FERMT1、LIPH、LAMA3及S100A16是胰腺癌的潜在诊断靶点及预后标志物,基于以上基因的预后模型表现出良好的预测效能。Objective To screen core methylation driver genes in pancreatic cancer,and to explore their biological functions and clinical significance.Methods Based on the mRNA-seq data of pancreatic cancer in TCGA/GEO database and the matched clinical data,using R analysis software related data packages and some online analysis tools,we analyzed and obtained the methylated driver genes in pancreatic cancer,and further screened the core methylated driver proteins by using Lasso-Cox regression analysis,and investigated their functional characteristics and clinical significance in pancreatic cancer.Finally,a prognostic model was constructed based on the core methylation driver genes and related investigations were carried out.Results Methylated driver genes ZSCAN18,FERMT1,LIPH,LAMA3 and S100A16 showed good diagnostic efficacy in pancreatic cancer,and the up-regulation of FERMT1,LIPH,LAMA3 and S100A16 was significantly correlated with poor prognosis of patients(P<0.05).Whereas the up-regulation of ZSCAN18 was correlated with better prognosis of patient survival(P<0.05).The prognostic model based on the methylation driver genes ZSCAN18,FERMT1,LIPH,LAMA3 and S100A16 showed good predictive efficacy and had good potential for clinical application.Conclusion Methylation driver genes ZSCAN18,FERMT1,LIPH,LAMA3 and S100A16 were potential diagnostic targets and prognostic markers for pancreatic cancer,and prognostic models based on these genes showed good predictive efficacy.
