摘　　要：目的:通过转录组测序技术(RNA-Seq)比较腹膜后脂肪肉瘤(RPL)组织与正常脂肪组织基因表达谱的差异,识别预后相关的关键基因并探究其潜在机制。方法:收集RPL患者肿瘤组织与瘤旁正常脂肪组织进行RNA-Seq并对差异表达基因进行GO和KEGG富集分析;基于TCGA数据库筛选预后相关的高风险基因并使用Kaplan-Meier曲线和受试者操作特征(ROC)曲线进行验证。结果:与正常脂肪组织相比,RPL组织中同时上调的基因共279个,主要富集在免疫反应过程和PPAR信号通路;结合TCGA数据集共鉴别出稳定的预后高风险基因7个,高风险组患者总生存率显著低于低风险组(P<0.05)。结论:KCNQ5、RBPJ等基因可能与RPL患者的不良预后相关,解析这些基因在RPL中的作用及机制有望为RPL患者诊疗策略的制定提供有力证据。Objective:To analyze and compare the differences of expression profiles between RPL and normal adipose tissue by transcriptome sequencing(RNA-Seq),then identify the key genes related to prognosis and explore their potential mechanisms.Methods:Tumor tissues and normal adipose tissues of patients with RPL were collected for RNA-Seq,and then the differentially expressed genes were analyzed by GO and KEGG enrichment analysis.Based on TCGA,the high-risk genes related to prognosis were screened and verified by Kaplan-Meier curve and receiver operating characteristic(ROC)curve.Results:Compared with normal adipose tissue,279 differentially expressed genes were simultaneously up-regulated in RPL tissues,which were mainly enriched in immune response and PPAR signaling pathway.Combined with TCGA,7 stable prognostic high risk genes were identified and the overall survival rate of the high risk group was significantly lower than that of the low risk group(P<0.05).Conclusion:KCNQ5,RBPJ and some other genes may be related to the poor prognosis of RPL patients.The analysis of the mechanism of these genes in RPL is expected to provide new evidence for the formulation of diagnosis and treatment strategies for RPL patients.
