摘  要:目的 探讨上调FSTL1对人肺腺癌细胞增殖、迁移的影响及可能分子机制。方法 将肺腺癌细胞株CL1-5分为对照组、10 ng/mL和30 ng/mL重组FSTL1组。CCK-8实验检测各组细胞的增殖能力;Transwell实验检测各组细胞的迁移能力;Western blot检测各组细胞的MMP2、MMP9和NF-κB/SPP1信号通路蛋白的表达。结果 FSTL1显著抑制CL1-5细胞增殖,且浓度越高对CL1-5细胞的增殖抑制作用越强;Transwell实验显示FSTL1可以明显抑制CL1-5细胞的迁移;FSTL1上调后,CL1-5细胞MMP2和MMP9的表达降低,NF-κB/SPP1信号通路被抑制。结论 FSTL1可能通过抑制NF-κB/SPP1信号通路抑制肺癌细胞的增殖和转移。Objective To elucidate the effects of FSTL1 on the proliferation and migration of human lung adenocarcinoma cells.Methods The experiment was divided into control group,10 ng/mL and 30 ng/mL recombinant FSTL1 group.Different concentrations of FSTL1 were used to treat CL1-5 cells.Cell proliferation was detected by CCK-8,and cell migration was detected by transwell test.Moreover,the expressions of MMP2,MMP9 and NF-κB/SPP1 were detected by western blot.Results FSTLlsignificantly inhibited the proliferation of CL1-5 cells,and the higher the concentration,the stronger the inhibition.FSTLlalso significantly decreased the migration of CL1-5 cells.After FSTL1 treatment,the expressions of MMP2 and MMP9 were significantly decreased in CL1-5 cells.Moreover,FSTL1 significantly decreased the expressions of NF-κBand SPP1.Conclusion FSTL1 can inhibit the proliferation and migration of CL1-5 cells,and its mechanism is related to theNF-κB/SPP1 signaling pathway.
