摘　　要：目的本研究从分子生物学水平探讨龙钻通痹方对类风湿关节炎(RA)外泌体miR-155的影响,以揭示龙钻通痹方如何干预其表达,从而为治疗RA夯实理论基础。方法本研究以胶原诱导型(CIA)的造模方式建立RA模型大鼠为研究对象,采用随机数字表法的方式将其分为正常组、模型组、龙钻通痹方组,经壮药龙钻通痹颗粒灌胃给药后,测定大鼠踝关节的肿胀指数;运用Elisa法检测血清肿瘤免疫因子白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)等炎症因子水平;并运用实时荧光定量PCR法检测血清外泌体中miR-155水平。结果不同组大鼠的踝关节肿胀程度不尽相同,正常组大鼠关节未见肿胀;而相较于治疗组,模型组大鼠未经治疗,关节肿胀程度更高(P<0.05)。治疗组大鼠血清IL-6、IL-1β、TNF-α水平均低于模型组(P<0.05)。治疗组大鼠血清外泌体miR-155水平高于正常组,但低于模型组(P<0.05)。结论壮药龙钻通痹方可调控miR-155的表达,抑制炎症因子的表达,进而发挥抗炎作用。Objective This study explores the effect of Longzuan Tongbi Formula on the exosome miR-155 in rheumatoid arthritis(RA)at the molecular biology level,in order to reveal the underlying mechanism and further consolidate the theoretical basis for the treatment of RA.Methods RA model was established by collagen-induced arthritis(CIA)modeling and the model rats were divided into the normal group,model group,and Longzuan Tongbi Formula group by the random number table method.The swelling index of the ankle joints of rats was measured after the gavage administration of Longzuan Tongbi Formula;the levels of serum tumor immune factors interleukin-6(IL-6),IL-1β,tumor necrosis factor(TNF)-α,and other inflammatory factors were measured by enzyme-linked immunosorbent assay(ELISA);and the level of miR-155 in serum exosomes was detected by real-time fluorescence quantitative PCR.Results The degree of ankle joint swelling varied among different groups of rats,and no joint swelling was observed in the normal group.Compared with the treated group,the untreated rats in the model group showed higher joint swelling(P<0.05).The serum levels of IL-6,IL-1βand TNF-αin the treated group were lower than those in the model group(P<0.05).The serum exosome miR-155 level in the treated group was higher than that in the normal group,but lower than that in the model group(P<0.05).Conclusion The Zhuang medicine Longzuan Tongbi Formula can regulate the expression of miR-155 and inhibit the expression of inflammatory factors,thereby exerting anti-inflammatory effects.
