摘  要:目的:对白术Atractylodes macrocephala Koidz.内生真菌米根霉Rhizopus oryzae的次生代谢产物进行研究。方法:利用大米蛋白胨固体培养基对米根霉Rhizopus oryzae进行发酵培养,利用多种柱色谱方法及制备高效液相等分离手段对该株真菌的次生代谢产物进行研究,根据多种波谱方法对分离得到的化合物进行结构鉴定。结果:从该次生代谢产物中分离纯化得到了26个化合物,分别鉴定为:麦角甾醇(1)、β-谷甾醇(2)、3β-O-acetyl-ergosterol(3)、3-acetate-ergosterol-5,8-endoperoxide(4)、(22E,24R)-ergosta-4,6,8(14),22-tetraen-3-one(5)、麦角甾-4,6,8(14),22-四烯-3-酮(6)、24-亚甲基环阿尔廷醇(7)、(3β,5α,22E)ergosta-6,8(14),22-triene-3,5-diol(8)、3β,5α-dihydroxy(22E,24R)-ergosta-7,22-dien-6-one(9)、chaxine C(10)、demethylincisterol A3(11)、吡咯-2-羧酸(12)、吲唑(13)、烟酸(14)、6-[(1R)-1-hydroxy-1-methylpropyl]-3-(2-methylpropyl)-2(1H)-pyrazinone(15)、pentadecanamide(16)、对羟基苯甲酸(17)、对羟基苯乙酸甲酯(18)、反式-阿魏酸(19)、对羟基苯甲醛(20)、对羟基苯乙酸(21)、辅酶Q9(22)、2,3-丁二醇(23)、9,9-dimethylhexacosane(24)、硬脂酸(25)、methyl(5Z,9Z,17R)-17-methyl-5,9-nonadecadienoate(26)。结论:以上化合物均为首次从米根霉Rhizopus oryzae中分离得到。Objective:To study the secondary metabolites of Rhizopus oryzae,an endophytic fungus from Atractylodes macrocephala.Methods:The Rhizopus oryzae was fermented in rice peptone medium.The secondary metabolites of Rhizopus oryzae were studied by column chromatography and HPLC.The structures of the compounds were identified by spectroscopic methods.Results:26 compounds were isolated from the secondary metabolites and identified as ergosterol(1),(-)-β-sitosterol(2),3β-O-acetyl-ergosterol(3),3-acetate-ergosterol-5,8-endoperoxide(4),(22E,24R)-ergosta-4,6,8(14),22-tetraen-3-one(5),ergosta-4,6,8(14),22-tetraen-3-one(6),24-methenecycloartenol(7),(3β,5α,22E)ergosta-6,8(14),22-triene-3,5-diol(8),3β,5α-dihydroxy(22E,24R)-ergosta-7,22-dien-6-one(9),chaxine C(10),demethylincisterol A3(11),pyrrole-2-carboxylic acid(12),indazole(13),nicotinic acid(14),6-[(1R)-1-hydroxy-1-methylpropyl]-3-(2-methylpropyl)-2(1H)-pyrazinone(15),pentadecanamide(16),4-hydroxybenzoic acid(17),methyl(4-hydroxyphenyl)acetate(18),trans-ferulic acid(19),4-hydroxybenzaldehyde(20),hydroxyphenylacetic acid(21),ubiquinone Q9(22),2,3-butanediol(23),9,9-dimethylhexacosane(24),stearic acid(25),methyl(5Z,9Z,17R)-17-methyl-5,9-nonadecadienoate(26),respectively.Conclusion:All compounds are isolated from Rhizopus oryzae for the first time.
