miR-29a-3p、FKBP14在宫颈癌组织中的表达及对预后的预测作用

摘　　要：目的探讨微小RNA-29a-3p(miR-29a-3p)、FK506相关蛋白14(FKBP14)在宫颈癌组织中的表达及对预后的预测作用。方法收集沧州市妇幼保健院2016年7月至2018年7月期间住院手术的110例宫颈癌患者的癌组织及癌旁组织,qRT-PCR检测miR-29a-3p表达水平,免疫组织化学检测FKBP14蛋白表达;miR-29a-3p与FKBP14表达的相关性采用Spearman相关分析;miR-29a-3p、FKBP14表达与预后的关系采用Kaplan-Meier法分析;多因素Cox回归分析患者预后的影响因素。结果与癌旁组织比较,宫颈癌组织中miR-29a-3p表达水平降低,FKBP14阳性率增加(P<0.05);低分化、肿瘤直径≥4cm、FIGO分期Ⅱ期miR-29a-3p低表达、FKBP14蛋白阳性表达患者比例高于中高分化、肿瘤直径<4 cm、FIGO分期Ⅰ期患者(P<0.05);宫颈癌组织miR-29a-3p与FKBP14的表达水平呈负相关(P<0.05);miR-29a-3p高表达患者5年生存率高于低表达患者,FKBP14阳性表达患者5年生存率低于阴性表达患者(P<0.05);FKBP14是患者死亡的危险因素,miR-29a-3p是保护因素(P<0.05)。结论宫颈癌组织miR-29a-3p低表达、FKBP14高表达,二者与患者5年预后有密切联系,有可能成为预后判断的生物指标。Objective To investigate the expression of microRNA-29a-3p(miR-29a-3p)and FK506-associated protein 14(FKBP14)in cervical cancer tissue and their predictive role in prognosis.Methods Cancer tissues and adjacent tissues of 110 patients with cervical cancer who were hospitalized and operated in Cangzhou Women and Children’s Health Hospital from July 2016 to July 2018 were collected.The expression level of miR-29a-3p was detected by qRT-PCR,and the expression of FKBP14 protein was detected by immunohistochemistry.The correlation between miR-29a-3p and FKBP14 expression was determined by Spearman correlation analysis.The relationship between the expression of miR-29a-3p and FKBP14 and prognosis was analyzed by Kaplan-Meier method.The influencing factors of prognosis were analyzed by Cox regression analysis.Results Compared with adjacent tissues,the expression level of miR-29a-3p in cervical cancer tissues was decreased,and the positive rate of FKBP14 increased(P<0.05).The proportion of patients with low miR-29a-3p expression and positive expression of FKBP14 protein in patients with low differentiation,tumor diameter≥4 cm and FIGO stageⅡwas higher than that in patients with medium and high differentiation,tumor diameter<4 cm and FIGO stageⅠ(P<0.05).The expression levels of miR-29a-3p and FKBP14 in cervical cancer tissues were negatively correlated(P<0.05).The 5-year survival rate of patients with high expression of miR-29a-3p was higher than that of patients with low expression,and the 5-year survival rate of patients with positive expression of FKBP14 was lower than that of patients with negative expression(P<0.05).FKBP14 was a risk factor for death,and miR-29a-3p was a protective factor(P<0.05).Conclusion The low expression of miR-29a-3p and high expression of FKBP14 in cervical cancer tissues are closely related to the 5-year prognosis of patients,and may become biological indicators of prognosis.
