摘  要:目的探讨南五灵复方不同提取物改善睡眠的功效。方法将120只雄性KM小鼠随机分为空白对照组(BCG)、醇提提取物组(EEG)、水提提取物组(WEG)、醇水提取物组(HEG),每组30只。各组小鼠灌胃相应剂量的药物,BCG组灌胃等量蒸馏水,每次0.1 ml/10 g,1次/d,连续给药30 d。末次给药后进行小鼠自主活动和直接睡眠实验、延长阈剂量戊巴比妥钠睡眠时间实验及阈下剂量戊巴比妥钠催眠实验,比较3种提取物对小鼠睡眠的影响。结果实验结果显示,各组小鼠末次给药后均未发生直接睡眠情况,同时各组小鼠自主活动情况比较差异无统计学意义(P>0.05),但各组小鼠睡眠潜伏期、睡眠时间比较差异均有统计学意义(均P<0.05)。与BCG组小鼠比较,EEG、HEG组小鼠睡眠潜伏期均缩短,睡眠时间均延长,同时HEG组延长小鼠睡眠时间效果优于WEG组,差异均有统计学意义(均P<0.05)。BCG组小鼠入睡率为20%(2/10),WEG组小鼠入睡率为30%(3/10),EEG组小鼠入睡率为60%(6/10),HEG组小鼠入睡率为80%(8/10);与BCG组小鼠比较,HEG组小鼠入睡率明显升高,差异有统计学意义(P<0.05)。结论南五灵复方醇提物和醇水提取物具有改善小鼠睡眠作用,醇水提取物改善睡眠的价值更高。Objective To investigate the efficacy of different extracts of Nanwuling Compound in improving sleep in mice.Methods One hundred and twenty male KM mice were randomized into a blank control group(BCG),an ethanol extract group(EEG),a water extract group(WEG),and a mixed extract group(HEG).Mice in each group were given corre⁃sponding doses of the drug by gavage,0.1 ml/10 g,once a day,for 30 consecutive days,except that BCG mice were given equal amount of distilled water by gavage.After the last administration,the mice were subjected to mouse spontaneous loco⁃motor activity and direct sleep experiment,prolonging the threshold dose of pentobarbital sodium sleep time experiment and subthreshold dose of pentobarbital sodium hypnosis experiment.The effects of the three extracts on the sleep status of mice were compared.Results The experimental results showed that no direct sleep occurred in any group after the last adminis⁃tration,and there was no significant difference in spontaneous locomotor activity between the groups(P>0.05).There were significant differences in sleep latency and sleep time between the groups(P<0.05).Compared with mice in the BCG group,mice in EEG and HEG groups had shorter sleep latency and longer sleep time(P<0.05).And the effect on prolon⁃ging sleep time in HEG group was better than that of the WEG group(P<0.05).The falling-asleep rates in mice in groups of BCG,WEG,EEG and HEG were 20%(2/10),30%(3/10),60%(6/10)and 80%(8/10),respectively.Compared with mice in the BCG group,the falling-asleep rate of mice in the HEG group significantly increased(P<0.05).Conclu⁃sion The ethanol extract and mixed extract of Nanwuling Compound have an effect on improving the sleep status in mice,and the effect of the latter is better.
