摘 要:目的探讨MAF1的表达与HCC不良预后之间的关联。方法挖掘TCGA、GTEx、HPA数据库中来建立MAF1表达与HCC之间的关联。利用ROC-AUC评估MAF1作为一个生物标志物的可靠性。通过qPCR验证MAF1在肝癌细胞中的相对表达水平。利用R工具GSVA的单样本基因集富集分析方法、TIMER数据库探索MAF1的表达与HCC中的免疫浸润水平之间的关系。结果果与正常对照组相比,HCC中MAF1存在高表达,且MAF1表达与不良预后之间呈现显著地正相关(P<0.05)。免疫浸润分析表明,MAF1的高表达与免疫细胞浸润和低生存率之间存在密切关联(P<0.05)。结论HCC组织中MAF1的高表达与不良预后相关,其机制可能是通过调控免疫细胞浸润促进癌症进展。MAF1可作为预测HCC预后的有价值的生物标志物。Objective To explore the association between MAF1 expression and adverse prognosis in hepatocellular carcinoma(HCC).Methods Data from TCGA,GTEx,and HPA databases were analyzed to establish the correlation between MAF1 expression and HCC.The reliability of MAF1 as a biomarker was assessed using ROC-AUC.qPCR was employed to validate the relative expression levels of MAF1 in liver cancer cells.The relationship between MAF1 expression and immune infiltration levels in HCC was explored using the single-sample gene set enrichment analysis(GSVA)method with R tool and the TIMER database.Results MAF1 exhibited higher expression in HCC compared to the normal control group,and its expression showed a significant positive correlation with adverse prognosis(P<0.05).Immune infiltration analysis indicated a close association between high MAF1 expression,immune cell infiltration,and low survival rates(P<0.05).Conclusion The high expression of MAF1 in HCC tissues is associated with adverse prognosis,and its mechanism may involve promoting cancer progression by regulating immune cell infiltration.MAF1 serves as a valuable biomarker for predicting the prognosis of HCC.
