摘  要:本文通过对月桂酰精氨酸乙酯(LAE)的抑菌谱、细菌细胞胞内的抑菌机制以及耐受消化道蛋白酶水解的研究发现,LAE对G+菌、G-菌、真菌均有广谱抑菌活性,且对G+菌的抑菌活性普遍高于G-菌和真菌。LAE在凝胶阻滞实验中可以与菌体基因组强烈结合,随浓度增加阻滞现象更明显。LAE能抑制胞内蛋白的合成,使得碱性磷酸酶、β-半乳糖苷酶表达活性降低。LAE对胃蛋白酶和胰蛋白酶的耐受能力都很差,胃蛋白酶可部分水解LAE,胰蛋白酶能完全水解LAE,证明LAE可被消化道内蛋白酶水解,不会在肠道内保留。The antibacterial spectrum of lauroyl arginate ethyl(LAE),its intracellular antibacterial mechanism and its tolerance to protease hydrolysis in gastrointestinal tract were studied.The results showed that LAE had broad-spectrum antibacterial activity against Gram-positive,Gram-negative,and antifungal activity.Its antibacterial activity against Gram-positive bacteria was generally higher than that against Gram-negative bacteria and fungi.The gel retardation assay showed that LAE could strongly bind to bacterial genome.This retardation phenomenon was more obvious with the increase of concentration of LAE.LAE could inhibit the synthesis of intracellular proteins and reduce the expression activities of alkaline phosphatase andβ-galactosidase.LAE had poor tolerance to pepsin and trypsin and could be partly hydrolyzed by pepsin and completely hydrolyzed by trypsin.It was proved that LAE could be hydrolyzed by gastrointestinal protease and could not be retained in the intestine.
