摘 要:为突破单菌培养在微生物类胡萝卜素生产中产率低的问题,采用5 L发酵罐等体积接种高活性粘红酵母和小球藻,研究菌藻混合培养工艺的可行性,进一步从转录水平分析混合体系特定时间的菌藻协同代谢分子机制。结果表明,混合培养可获得最大生物量、残糖利用率、类胡萝卜素产量分别为51.7 g/L、83.3%和29.8 mg/L,实验组类胡萝卜素产量比菌藻纯培养组分别提高了21.5%和77.2%,且混合培养可获得7种类胡萝卜素,提高了玉米黄质和虾青素的占比;发酵36 h时取样测定分析转录组数据发现,实验组与菌藻对照组对比分别获得85个和2858个差异表达基因,GO聚类和KEGG代谢通路分析表明,小球藻可通过强化光合作用和特定类胡萝卜素合成相关基因的上调,实现兼养和改变类胡萝卜素种类的占比。A mixed culture was used in producing natural carotenoids to overcome the problem of low production rate in single microbe culture.An equal volume of Rhodotorula glutinis and Chlorella vulgaris was inoculated in a 5 L fermentation tank.The feasibility of the mixed culture process of bacteria and algae was studied,the molecular mechanism of synergistic metabolism between R.glutinis and C.vulgaris in the mixed system was analyzed at transcriptional level at specific time.The results showed that the maximum biomass,residual sugar utilization rate and carotenoids yield of mixed culture were 51.7 g/L,83.3%and 29.8 mg/L,respectively.The carotenoids yield of experimental group were 21.5%and 77.2%higher than those of the single culture groups of R.glutinis or C.vulgaris,respectively.Moreover,the mixed culture could obtain seven kinds of carotenoids and proportion of zeaxanthin and astaxanthin was also increased.Samples were taken at 36 h of fermentation,and transcriptome sequencing analysis showed that 85 and 2858 genes were obtained in the experimental and the control groups,respectively.Gene ontology classification and KEGG metabolic pathway analysis indicated that C.vulgaris had dual function of obtaining the nutrition and adjusting proportion of carotenoid species by enhancing upregulation genes related to photosynthesis and carotenoids synthesis.
