摘  要:目的:基于癌症基因组图谱(The Cancer Genome Atlas,TCGA)数据集,探索tspear基因在结直肠癌中的预后作用及机制。方法:自TCGA获取数据,应用R语言分析结直肠癌组织和正常组织间tspearmRNA的表达差异及TSPEAR表达与临床病理特征之间的相关性。采用Kaplan-Meier分析、比例风险回归分析(Cox's proportional hazards regression model,Cox)和列线图确定tspear对结直肠癌患者总生存率和疾病特异性生存率的预测价值。对tspear相关的差异表达基因进行基因集富集分析(Gene Set Enrichment Analysis,GSEA)、基因本体富集(Gene Ontology,GO)和信号通路富集(Kyoto Encyclopedia of Genes and Genomes,KEGG)。结果:tspear在结直肠癌中的表达显著增加,其表达升高与肿瘤T分期、M分期、残余肿瘤、总生存期和疾病特异性生存期显著相关。单变量Cox回归分析表明tspear是结直肠癌生存率的预测因子。多变量Cox回归分析显示tspear是结直肠癌的独立预后因素。KEGG分析显示tspear相关差异表达基因主要富集于神经活性配体-受体相互作用通路。结论:tspear可能是一种新的结直肠癌独立预后生物标志物。Objective:To explore the prognostic role and mechanism of TSPEAR in colorectal cancer(CRC)based on The Cancer Genome Atlas(TCGA)data set.Methods:Based on TCGA data,the expression difference of tspear mRNA between CRC and normal tissues,and the correlation between TSPEAR expression and clinic pathological features were analyzed by R language.Kaplan-Meier analysis,Cox regression analysis,and nomograms were used to determine the predictive value of TSPEAR on overall survival(OS)and disease-specific survival(DSS)in CRC patients.TSPEAR-related differentially expressed genes(DEGs)were screened and analyzed by Gene Set Enrichment Analysis(GSEA),Gene Ontology(GO),and Kyoto Encyclopedia of Genes and Genomes(KEGG).Results:TSPEAR expression increased significantly in CRC,which was significantly correlated with T stage,M stage,residual tumor,OS event,and DSS event.Univariate Cox regression analysis indicated that TSPEAR was a predictor of OS and DSS.Multivariate Cox regression analysis indicated that TSPEAR was an independent prognostic factor for CRC.Functional annotation indicated that TSPEAR-related DEGs were enriched in neuroactive ligand-receptor interaction.Conclusion:TSPEAR may be a novel independent prognostic biomarker for CRC.
