摘 要:细胞焦亡是一种炎性相关的细胞程序性死亡方式,其特征是由炎性小体,半胱天冬氨酸蛋白酶以及消皮素蛋白家族介导发生,导致胞膜形成微孔,细胞肿胀破裂,进而释放大量的炎症因子以及细胞内容物。大量研究证实细胞焦亡参与了酒精性肝病(Alcoholic liver disease,ALD),非酒精性脂肪性肝病(Non-alcoholic fatty liver disease,NAFLD)和肝纤维化等多种肝脏疾病的发病过程。当前,基于细胞焦亡学说如何有效防治肝脏疾病成为该领域新的研究热点。天然化合物来源广泛,具有多靶点、多通路、多药效等特点,是治疗肝脏疾病的重要物质来源。该文简要概述了细胞焦亡发生的分子机制和焦亡与肝脏疾病的关系,对近年来天然化合物靶向细胞焦亡改善肝脏疾病的研究进行归纳总结,以期为临床开发新药治疗肝脏疾病提供最新的思路和方向。Pyrodeath is an inflammatory process of programmed cell death.It is mediated by inflammasome,casPASE protease,and dermatin protein families,resulting in the formation of micropores in the cell membrane,cell swelling and rupture,and the release of a large number of inflammatory factors and cell contents.A large number of studies have confirmed that pyrodeath is involved in the pathogenesis of many liver diseases such as Alcoholic liver disease(ALD),Non-alcoholic fatty liver disease(NAFLD)and liver fibrosis.At present,how to effectively prevent liver diseases based on the theory of pyrodeath has become a new research hotspot in this field.Natural compounds come from a wide range of sources,with the characteristics of multi-target,multi-pathway,multi-efficacy,etc.,and are important material sources for the treatment of liver diseases.In this paper,the molecular mechanism of pyrodeath and the relationship between pyrodeath and liver disease were briefly reviewed,and the recent researches on the improvement of liver disease by natural compounds targeting pyrodeath were summarized,in order to provide the latest ideas and directions for clinical development of new drugs for liver disease.
