摘 要:脑胶质瘤是起源于神经中枢系统(CNS)恶性程度最高且伴随高死亡率与致残率的原发性肿瘤。DNA甲基转移酶1(DNMT1)是与脱氧核糖核苷酸甲基化作用关系密切的关键酶,在脑胶质瘤中的异常表达与肿瘤的发生发展密切相关,其高表达可促进脑胶质瘤生物学功能增强及耐药性产生。此外,DNMT1可调控不同基因甲基化的表观遗传学机制也为脑胶质瘤的治疗提供了新靶点,但其治疗效果因其泛甲基化的特点而具有争议。本文旨在对近年来DNMT1在脑胶质瘤中的研究进行综述,系统分析DNMT1在脑胶质瘤发生、治疗及预后中的作用,为脑胶质瘤治疗提供参考。Glioma stands as the most aggressive primary tumor arising within the central nervous system(CNS)and is associated with high mortality and disability rates.DNA methyltransferase 1(DNMT1),a key enzyme closely related to DNA methylation,is significantly associated with the occurrence and progression of glioma.Its overexpression has been found to be related to the invasiveness,deterioration,and drug resistance of glioma.And it was positively correlated with the proliferation and metastasis of glioma.Moreover,DNMT1’s involvement in regulating the expression of various genes offers multiple potential targets for glioma treatment.This review provides an overview of the research progress on DNMT1 in glioma over the past decade,offering a systematic analysis of its role in glioma development,treatment,and prognosis.The aim is to present new perspectives and strategies for glioma treatment.
