摘 要:目的观察汉黄芩素对青光眼小鼠视网膜神经节细胞(RGC)的保护作用,并探究其相关作用机制。方法10只(10眼)C57BL/6J小鼠作为对照组;40只(40眼)DBA/2J小鼠用于制备青光眼模型,并分为模型组和实验1、2、3组,每组各10只。对照组、模型组小鼠灌胃生理盐水,实验1、2、3组小鼠分别灌胃50、100、150 mg·kg^(-1)汉黄芩素。以小鼠右眼为入选眼,比较各组小鼠灌胃前和灌胃2周、4周、8周眼压;苏木素-伊红染色并比较各组小鼠RGC数量、视网膜厚度;比色法检测各组小鼠视网膜组织超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、一氧化氮合酶(NOS)水平;酶联免疫吸附法检测各组小鼠视网膜组织白细胞介素(IL)-6、IL-1β、肿瘤坏死因子-α(TNF-α)水平;Western blot检测各组小鼠视网膜组织核苷酸寡聚化结构域样受体家族3(NLRP3)、半胱天冬酶-1(Caspase-1)、凋亡相关斑点样蛋白(ASC)的蛋白表达水平。结果灌胃前,与对照组相比,模型组和实验1、2、3组小鼠眼压均升高,差异均有统计学意义(均为P<0.05)。灌胃2周、4周、8周:与对照组相比,模型组和实验1、2、3组小鼠眼压均增加,差异均有统计学意义(均为P<0.05);与模型组相比,实验1、2、3组小鼠眼压均降低,差异均有统计学意义(均为P<0.05);与实验1组相比,实验2、3组小鼠眼压均降低,差异均有统计学意义(均为P<0.05);与实验2组相比,实验3组小鼠眼压均降低,差异均有统计学意义(均为P<0.05)。与对照组相比,模型组和实验1、2、3组小鼠RGC数量及视网膜厚度、SOD水平、GSH-Px水平均降低,NOS、IL-6、IL-1β、TNF-α水平以及NLRP3蛋白、Caspase-1蛋白、ASC蛋白表达水平均增加,差异均有统计学意义(均为P<0.05);与模型组相比,实验1、2、3组小鼠RGC数量及视网膜厚度、SOD水平、GSH-Px水平均增加,NOS、IL-6、IL-1β、TNF-α水平以及NLRP3蛋白、Caspase-1蛋白、ASC蛋白表达水平均降低,差Objective To explore the protective effect of wogonin on retinal ganglion cells(RGC)in mice with glaucoma and its related mechanism.Methods Ten(10 eyes)C57BL/6J mice were included in the control group,and 40(40 eyes)DBA/2J mice were selected to prepare glaucoma models,which were divided into the model group,experiment 1,2 and 3 groups,with 10 mice per group.Mice in the control and model groups were given normal saline intragastrically,and mice in the experiment 1,2 and 3 groups were intragastrically given 50,100 and 150 mg·kg^(-1)wogonin,respectively.The right eyes were selected as experimental eyes.The intraocular pressure(IOP)of mice was compared among the five groups before and after 2,4 and 8 weeks of intragastric administration.The RGC number and retinal thickness of mice in each group were compared after hematoxylin-eosin staining.The levels of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)and nitric oxide synthase(NOS)in the retinal tissues of mice were detected by colorimetry.The levels of interleukin(IL)-6,IL-1βand tumor necrosis factor-α(TNF-α)in the retinal tissues of mice were detected by enzyme-linked immunosorbent assay.The expression levels of nucleotide oligomerization domain-like receptor family 3(NLRP3),Caspase-1 and apoptosis-associated speck-like protein(ASC)in the retinal tissues of mice were detected by Western blot.Results Compared with the control group,IOP in the model group and the experiment 1,2 and 3 groups increased(all P<0.05)before intragastric administration.After 2,4 and 8 weeks of intragastric administration,IOP in the model group and the experiment 1,2 and 3 groups increased compared with the control group(all P<0.05);IOP in the experiment 1,2 and 3 groups decreased compared with the model group(all P<0.05);IOP in the experiment 2 and 3 groups decreased compared with the experiment 1 group(both P<0.05);IOP in the experiment 3 group decreased compared with the experiment 2 group(P<0.05).Compared with the control group,the number of RGC,retinal thickness,SOD and GSH-
