摘  要:中药分析学是中药制药专业学生的核心专业课程。掌握中药多糖的制备与表征是中药分析学实验教学的重要部分。本文以常用中药黄连为研究对象,设计黄连多糖的制备与表征综合实验。首先采用水提醇沉的方法制备黄连多糖,之后对黄连多糖进行紫外-可见和红外光谱表征,最后明确黄连多糖的分子量分布及单糖组成。该实验能够全面培养学生的实验能力,包括多糖的制备技术、光谱解析、高效液相色谱仪的使用以及样品的柱前衍生化处理技术等。通过实验教学改革,激发学生的学习兴趣,提高学生的科研与创新能力。Traditional Chinese medicine analysis is the core course for students majoring in Chinese pharmaceutical manufacturing.Mastering the preparation and characterization of traditional Chinese medicine polysaccharides is an important part of experimental teaching of traditional Chinese medicine analysis.In this paper,Coptis Chinensis,a commonly used traditional Chinese medicine was selected as the research object and a comprehensive experiment about the preparation and characterization of Coptis Chinensis polysaccharides was designed.First,Coptis Chinensis polysaccharides were prepared with water extraction and alcohol precipitation method.Next,the Coptis Chinensis polysaccharides were characterized by UV-Vis and IR spectroscopy.Finally,the molecular weight distribution and monosaccharide composition was clarified.This experiment could comprehensively cultivate students’experimental ability,including the preparation of polysaccharides,the analysis of spectral,the use of high-performance liquid chromatography,and the precolumn derivatization technique.Through experiment teaching reform,the students’interest could be stimulated,and students’ability of scientific research and innovation could be improved.
