摘 要:目的:探究膜相关环指蛋白7(MARCH7)在上皮性卵巢癌(EOC)组织和细胞中的表达及作用机制。方法:选取就诊且经手术治疗的患者32例,包括EOC患者16例和卵巢组织正常患者16例,收集EOC患者和卵巢组织正常患者的卵巢组织标本,采用实时荧光定量PCR(RT-qPCR)检测MARCH7 mRNA表达水平。构建过/低表达MARCH7的上皮性卵巢癌细胞(SKOV3、CAOV3)后,采用Transwell实验观察MARCH7对EOC细胞迁移和侵袭能力的影响,采用Western blot检测过/低表达MARCH7的EOC细胞上皮-间质转化(EMT)相关蛋白E-钙粘蛋白(E-cadherin)、N-钙粘蛋白(N-cadherin)、锌指转录因子1(Snail1)、锌指转录因子2(Slug)蛋白表达。结果:卵巢癌组织MARCH7 mRNA表达水平高于正常卵巢组(P<0.05)。在SKOV3和CAOV3细胞中,正常对照组与低表达对照组和过表达对照组细胞迁移和侵袭能力,以及MARCH7和EMT相关蛋白E-cadherin、N-cadherin、Snail1和Slug蛋白表达比较无统计学差异(均P>0.05);与低表达对照组比较,沉默MARCH7的细胞迁移和侵袭能力降低,E-cadherin蛋白表达升高,N-cadherin、Snail1和Slug蛋白表达降低(均P<0.01);与过表达对照组比较,过表达MARCH7的细胞迁移和侵袭能力升高,E-cadherin蛋白表达降低,N-cadherin、Snail1和Slug蛋白表达升高(均P<0.01)。结论:MARCH7在EOC组织和细胞中呈高表达,可能通过促进EOC细胞的EMT过程,增强EOC细胞的侵袭和迁移能力。Objective:To investigate the expression and mechanism of membrane-associated ring finger protein 7(MARCH7)in epithelial ovarian cancer(EOC)tissues and cells.Methods:A total of 32 patients who underwent surgical treatment were enrolled in this study,including 16 patients with EOC and 16 patients with normal ovarian tissues.Ovarian tissues from EOC patients and 16 patients with normal ovarian tissues were collected,and the expression of MARCH7 mRNA was detected by RT-qPCR.After ovarian cancer cells(SKOV3,CAOV3)with overexpression or knockdown of MARCH7 were constructed,Transwell assay was used to observe the effect of MARCH7 on the migration and invasion of EOC cells,and Western blot was used to detect EMT-related proteins E-cadherin,N-cadherin,Snail1 and Slug in MARCH7 overexpressed/down-regulated EOC cells.Results:The expression of MARCH7 mRNA in ovarian cancer tissues was significantly higher than that in normal ovarian tissues(P<0.05).In SKOV3 and CAOV3 cells,there was no significant difference in the migration and invasion ability between the normal control group and the low expression control group and the overexpression control group,as well as the expression of MARCH7 and EMT related proteins E-cadherin,N-cadherin,Snail1 and Slug(all P>0.05);compared with the low expression control group,the migration and invasion abilities of MARCH7 knockdown cells were decreased,the expression of E-cadherin protein was increased,and the expressions of N-cadherin,Snail1 and Slug protein were decreased(all P<0.01);compared with the overexpression control group,the migration and invasion abilities of MARCH7 overexpression cells were increased,the expression of E-cadherin protein was decreased,and the expressions of N-cadherin,Snail1 and Slug protein were increased(all P<0.01).Conclusion:MARCH7 is highly expressed in EOC tissues and cells,which may promote the invasion and migration of EOC cells by promoting the EMT process.
