摘 要:为探究化学修饰对雨生红球藻多糖(Haematococcus pluvialis polysaccharide,HPP)体外抗肿瘤活性的影响,从雨生红球藻提取得到HPP,分别对其进行羧甲基化、硫酸化和乙酰化修饰,对获得的修饰多糖进行体外抗肿瘤活性的研究。对HPP进行羧甲基化修饰得到C-HPP,其取代度为0.17;对HPP进行硫酸化修饰得到S-HPP,其取代度为0.22;对HPP进行乙酰化修饰得到A-HPP,其取代度为0.25。抗肿瘤试验结果表明,C-HPP、S-HPP和A-HPP体外抑制肝癌细胞Hep G-2和宫颈癌细胞Hela增殖的效果均优于HPP,其中乙酰化修饰对2种细胞增殖的抑制效果最好,IC_(50)分别为0.637 mg/mL和0.867 mg/mL。综上所述,对雨生红球藻多糖进行化学修饰可以提高其体外抑制肝癌细胞Hep G-2和宫颈癌细胞Hela增殖的活性,研究结果可为雨生红球藻多糖的进一步开发利用提供依据。In order to explore the effect of chemical modification on the antitumor activity of Haematococcus pluvialis polysaccharide(HPP) in vitro,HPP was extracted from Haematococcus pluvialis and modif ied by c arbox ymethylation,sulfation and acet ylation,respectively.The antitumor activity of the modified polysaccharide in vitro was studied.C-HPP was obtained by carboxymethylation of HPP with a degree of substitution of 0.17.HPP was sulfated to obtain S-HPP with a degree of substitution of 0.22.HPP was acetylated to obtain A-HPP with a degree of substitution of 0.25.The results of anti-tumor test showed that C-HPP,S-HPP and A-HPP were superior to HPP in inhibiting the proliferation of HepG-2 and Hela in vitro.Among them,acetylation modification had the best inhibitory effect on the proliferation of the two cells,with IC_(50) values of 0.637 mg/mL and 0.867 mg/mL,respectively.In summary,the chemical modification of HPP can improve its activity of inhibiting the proliferation of hepatoma cell HepG-2 and cervical cancer cell Hela in vitro.The results provide a basis for the further development and utilization of HPP.
