八子补肾胶囊延缓SAMP6小鼠增龄性胸腺退化的初探

摘  要:目的探讨八子补肾胶囊对SAMP6小鼠胸腺退化的改善作用及其可能机制。方法20只12周龄雄性SAMP6小鼠随机分为模型组(SAMP6)和八子补肾治疗组(SAMP6+BZBS),10只SAMR1小鼠设为同源对照组(SAMR1)。SAMP6+BZBS组每日灌胃八子补肾胶囊混悬液(2.8 g·kg^(-1)),其余两组每日灌胃等量蒸馏水,给药9周后观察各组胸腺形态并计算胸腺指数;HE染色观察胸腺组织结构变化;SA-β-gal染色检测胸腺衰老情况;流式细胞术检测各组胸腺CD3+T细胞比例;Western blot检测胸腺中p16、Bax、Bcl-2、cleaved caspase-3蛋白水平;免疫荧光检测各组皮质胸腺上皮细胞比例;ELISA检测胸腺中IL-7水平。结果与SAMP6组小鼠比较,SAMP6+BZBS组胸腺指数明显提高(P<0.05);紊乱的胸腺结构得到明显改善;SA-β-gal染色阳性比例明显降低(P<0.01);CD3+T细胞比例明显提高(P<0.05);p16蛋白水平明显降低(P<0.05);Bcl-2蛋白水平明显增加(P<0.05),而cleaved caspase-3蛋白水平明显减少(P<0.05);皮质胸腺上皮细胞比例明显提高;IL-7水平明显升高(P<0.01)。结论八子补肾胶囊能够延缓SAMP6小鼠的胸腺退化、抑制胸腺中细胞凋亡、有助于胸腺细胞发育,可能与其提高皮质胸腺上皮细胞比例,促进IL-7分泌相关。Aim To explore the improvement effect of Bazi Bushen capsule on thymic degeneration in SAMP6 mice and the possible mechanism.Methods Twenty 12 week old male SAMP6 mice were randomly divided into the model group(SAMP6)and the Bazi Busheng capsule treatment group(SAMP6+BZBS).Ten SAMR1 mice were assigned to a homologous control group(SAMR1).The SAMP6+BZBS group was orally administered Bazi Bushen capsule suspension(2.8 g·kg^(-1))daily,while the other two groups were orally administered an equal amount of distilled water.After nine weeks of administration,the morphology of the thymus in each group was observed and the thymus index was calculated;HE staining was used to observe the structural changes of thymus tissue;SA-β-gal staining was used to detect thymic aging;flow cytometry was used to detect the proportion of thymic CD3+T cells in each group;Western blot was used to detect the levels of p16,Bax,Bcl-2,and cleaved caspase-3 proteins in thymus;immunofluorescence was applied to detect the proportion of cortical thymic epithelial cells in each group;ELISA was employed to detect IL-7 levels in thymus.Results Compared with the SAMP6 group,the thymic index of the SAMP6+BZBS group significantly increased(P<0.05);the disordered thymic structure was significantly improved;the positive proportion of SA-β-gal staining significantly decreased(P<0.01);the proportion of CD3+T cells apparently increased(P<0.05);the level of p16 protein significantly decreased(P<0.05);the level of Bcl-2 protein significantly increased(P<0.05),while the level of cleaved caspase-3 protein markedly decreased(P<0.05);the proportion of cortical thymic epithelial cells evidently increased;the level of IL-7 significantly increased(P<0.01).Conclusions Bazi Bushen capsule can delay thymic degeneration,inhibit cell apoptosis in thymus and promote thymic cell development in SAMP6 mice,which may be related to increasing the proportion of cortical thymic epithelial cells and promoting IL-7 secretion.
