摘 要:建立符合随行标准汤剂要求的肉桂配方颗粒制备方法和特征图谱。利用挥发油和水提液明确挥发性成分和水溶性成分,收集肉桂标准汤剂及肉桂配方颗粒各环节的样品的特征图谱中挥发性成分和水溶性成分的比值(H),以肉桂标准汤剂中该比值H汤为参照并利用该比值对肉桂配方颗粒制备过程中挥发油包合物及浓缩液进行调配.UPLC特征图谱确定了5个特征峰,其中1~4号峰分别为原儿茶酸、香豆素、肉桂酸、桂皮醛,并确认了桂皮醛为挥发性成分,其余各成分为水溶性成分;根据挥发性成分和水溶性成分的比值关系,在明确了包合率、包合物得率的基础上,得到了挥发油包合物与浓缩液的调配公式,该公式调配后获得的肉桂配方颗粒的H比值为9.25,与随行肉桂标准汤剂的H比值9.08接近,实现了肉桂配方颗粒与肉桂标准汤剂化学成分一致.首次利用特征图谱建立了肉桂配方颗粒制备过程中挥发油包合物与浓缩液的调配方法,该方法操作简单,可保证生产出的配方颗粒在挥发性成分和水溶性成分比例的一致性,对肉桂配方颗粒的生产具有指导意义和参考价值.This study is to establish a preparation method of Cinnamomum cassia formula granules which meets the requirements of accompanying standard decoction.The characteristic map of Cinnamon standard decotion and formu⁃la particles was established.The volatile components and water⁃soluble components were identified by volatile oil and water extract.The ratio of volatile components to water⁃soluble components in the characteristic atlas of Cinna⁃momum cassia standard decoction and Cinnamomum cassia formula granules was collected.Taking the ratio H in Cinnamomum cassia Standard decoction as reference and using the ratio,the volatile oil inclusion compound and concentrated solution in the preparation process of Cinnamomum cassia formula particles were mixed.Five charac⁃teristic peaks were determined by UPLC characteristic map,among which the peaks 1~4 were protocatechuic acid,coumarin,cinnamic acid and cinnamaldehyde,respectively.Cinnamaldehyde was confirmed as volatile component,and the other components were divided into water⁃soluble components.According to the ratio relationship between volatile components and water⁃soluble components,the blending formula of volatile oil inclusion complex and con⁃centrated solution was established on the basis of defining the inclusion rate and the yield rate of the inclusion com⁃plex.The H ratio of Cinnamon formula particles prepared by this formula is consistent with that of Cinnamomum cas⁃sia standard decoction.For the first time,the blending method of volatile oil inclusion compound and concentrated solution in the preparation of Cinnamon formula particles was established by using characteristic atlas for the first time.The method is simple to operate,can ensure the consistency of volatile components and water⁃soluble compo⁃nents,and has guiding significance and reference value for the production of Cinnamomum cassia formula particles.
