摘  要:目的研究N-甲基-D-天冬氨酸受体(NMDA)亚型NMDAR1、NR2A和NR2B在尼古丁戒断大鼠海马组织中的表达及观察焦虑样行为。方法20只健康雄性SD大鼠按随机数字表法分为正常组和模型组,每组10只,适应性单笼饲养1周后开始建模。模型组大鼠连续7 d给尼古丁(0.4 mg/kg/次,2次/d)皮下注射,撤药72 h,建立尼古丁戒断模型。正常对照组大鼠连续7 d给生理盐水(1 mL/次,2次/d)皮下注射。采用高架十字迷宫检测两组大鼠焦虑样行为,以判断模型制备是否成功。行为学检测结束后,将大鼠异氟烷麻醉后,处死大鼠,取海马组织备用。采用qRT-PCR技术检测两组大鼠海马组织NMDAR1、NR2A和NR2B的mRNA表达,采用WB技术检测两组大鼠海马组织NMDAR1、NR2A和NR2B的蛋白表达。结果尼古丁戒断模型组大鼠明显焦虑,与正常组相比,模型组大鼠进入开放臂的次数、开放臂滞留时间、开放臂进入次数百分比、开放臂滞留时间百分比都显著降低(P<0.05),总路程无统计学差异,提示模型制备成功;qRT-PCR检测结果显示,与正常组相比,模型组大鼠海马组织中NMDAR1、NR2A和NR2B的mRNA表达显著降低(P<0.05);WB结果显示,与空白对照组相比,模型组大鼠海马组织内NMDAR1、NR2A和NR2B的蛋白相对表达水平显著降低(P<0.05)。结论尼古丁戒断大鼠出现行为敏感化,NMDA受体亚型NMDAR1、NR2A和NR2B表达异常可能与尼古丁戒断后的焦虑样行为密切相关。Objective To study the expression of N-methyl-D-aspartate receptor(NMDA)subtypes NMDAR1,NR2A,and NR2B in the hippocampal tissues of nicotine withdrawal rats and to observe anxiety-like behavior.Methods Twenty healthy male SD rats were divided into normal and model groups according to the random number table method,with 10 rats in each group,and the modeling was started after 1 week of acclimatized single-cage rearing.Rats in the model group were administered nicotine(0.4 mg/kg/dose,2 times/day)subcutaneously for 7 consecutive,and the drug was withdrawn for 72 h to establish a nicotine withdrawal model.Normal control rats were injected with saline(1 mL/time,2 times/day)subcutaneously for 7 consecutive days.Anxiety-like behaviors of rats in both groups were detected by using an elevated cross maze to determine whether the model preparation was successful or not.After the behavioral detection,the rats were anesthetized with isoflurane,then the rats were executed and the hippocampal tissues were taken for spare.The mRNA expression of NMDAR1,NR2A,and NR2B in the hippocampal tissues of the two groups was detected by qRT-PCR,and the protein expression of NMDAR1,NR2A,and NR2B in the hippocampal tissues of the two groups was detected by Western Blot.Results The rats in the nicotine withdrawal model group were anxious.Compared with the normal group,times of entered the open arm,the duration of staying in the open arm,the percentage of entering the open arm,and the percentage of staying in the open arm were significantly lower in the model group(P<0.05),and there was no difference in the total distance traveled,suggesting that the model preparation was successful.The qRT-PCR results showed that NMDAR1,NR2A,and NR2B in the hippocampal tissue of rats in the model group(P<0.05).Western Blot results showed that the relative protein expression levels of NMDAR1,NR2A,and NR2B in the hippocampal tissue of rats in the model group were significantly lower than those in the blank control group(P<0.05).Conclusion Behavioral sensitizati
