摘 要:目的探讨氟马替尼、地西他滨、三氧化二砷以及维奈托克对人慢性髓系白血病(chronic myeloid leukemia,CML)K562细胞系增殖的影响,为CML新的治疗方案提供实验室依据。方法利用不同浓度的氟马替尼、地西他滨、三氧化二砷、维奈托克分别单独及两药联合作用于K562细胞,采用CCK-8法测定细胞增殖活力,采用Compusyn软件计算两药联合作用的联合指数。结果不同浓度的氟马替尼、三氧化二砷、地西他滨及维奈托克单药均可抑制K562细胞增殖,差异均有统计学意义(P<0.01);氟马替尼、地西他滨、三氧化二砷分别两药联合可增强对K562细胞的抑制作用(P<0.01),但分别与维奈托克联合后对K562细胞的抑制无明显增强(P>0.05)。结论氟马替尼、三氧化二砷、地西他滨、维奈托克单药均能抑制K562细胞的增殖,氟马替尼、地西他滨、三氧化二砷分别两药联合均有协同作用;而这3种药物分别与维奈托克联合无明显协同作用。Objective To explore the effects of flumatinib,decitabine,arsenic trioxide and venetoclax on the proliferation of K562 cell line in human chronic myeloid leukemia(CML),and provide laboratory basis for new treatment options for CML.Methods Different concentrations of flumatinib,decitabine,arsenic trioxide,and venetoclax were used to treat on K562 cells alone or in combination.The cell proliferation activity was determined by CCK8method,and the combined index of the two drugs was calculated by Compusyn software.Results Different concentrations of flumatinib,arsenic trioxide,decitabine and venetoclax alone could inhibit the proliferation of K562 cells,and the differences were statistically significant(P<0.01);The combination of two drugs with flumatinib,decitabine and arsenic trioxide could enhance the inhibition effect on K562 cells(P<0.01),but the combination with venetoclax was not significantly enhancement on the inhibition effect on K562 cells(P>0.05).Conclusion Flumatinib,arsenic trioxide,decitabine,and venetoclax alone can inhibit the proliferation of K562 cells,and the combination of two drugs with flumatinib,decitabine,and arsenic trioxide has synergistic effect.There was no obvious synergistic effect between the three drugs combined with venetoclax,respectively.
