摘 要:为了鉴定优异育种材料的Waxy(Wx)基因型及其相关性状的表型,利用DNA分子标记,结合特异引物扩增片段测序,对27份水稻优异育种材料Wx基因第1内含子上游的(CT)n序列、第1内含子+1位G/T单核苷酸突变(single nucleotide polymorphism,SNP)、第4外显子A/G SNP和第6外显子的A/C SNP进行了鉴定;同时对直链淀粉含量(amylose content,AC)和胶稠度(gel consistency,GC)进行了测定。结果表明,27份水稻材料Wx基因第1内含子上游共有5种(CT)n多态性(n=10,11,12,17,18),包括Wxa型6份、Wxb型16份、Wxhp型3份、Wxin型1份和wx型1份;上述5种Wx基因型水稻对应的AC含量分别为23.1%~29.0%、10.9%~16.4%、5.7%~9.8%、18.4%和3.5%,(CT)n多态性和G/T SNP分别能解释88.9%和85.2%的直链淀粉含量变异,且(CT)n多态性标记具有较高的选择准确性;AC与GC间呈显著的负相关(r=-0.4827*),但也存在高直高胶和低直低胶的特异材料。研究结果对培育优良杂交稻亲本和组配优质杂交稻品种具有指导意义。In order to identify the Wx genotype and the phenotype of its related traits in the elite breeding materials,the(CT)n sequence upstream of the first intron of the Wx gene,the first intron+1 G/T SNP,the A/G SNP of exon 4 and the A/C SNP of exon 6 of 27 elite rice breeding materials were identified by DNA molecular markers combined with specific primer amplification fragment sequencing.Amylose content(AC)and gel consistency(GC)were also measured.The results showed that there were five(CT)n polymorphisms(n=10,11,12,17,18)upstream of the first intron of the Wx gene in the 27 rice materials.Six of them were of the Wxa genotype,16 were of the Wxb genotype,three were of the Wxhp genotype,one was of the Wxin genotype,and one was wx.Wxa materials had higher AC(23.1%~29.0%),Wxb materials had moderate AC(10.9%~16.4%),Wxhp materials had lower AC(5.7%~9.8%),Wxin materials had an AC of 18.4%,and wx materials were glutinous rice with an AC of 3.5%.(CT)n polymorphisms and G/T SNP explained 88.9%and 85.2%of the variation in amylose content,respectively,and(CT)n polymorphism markers had high selection accuracy.There was a significant negative correlation between AC and GC(r=-0.4827*),but there were also specific materials with high AC and high GC,or low AC and low GC.This study provides material and technical references for cultivating excellent hybrid rice parents,breeding high-quality and special hybrid rice varieties.
