摘 要:目的:研究三七皂苷Ft1对宫颈癌Siha细胞迁移、侵袭及凋亡的影响及作用机制。方法:取对数生长期宫颈癌Siha细胞,将Siha细胞分为对照组(0μmol/L)、低浓度组(20μmol/L)、中浓度组(40μmol/L)、高浓度组(80μmol/L),采用CCK-8法检测细胞增殖活性,以平板克隆实验、细胞划痕实验和Transwell实验检测三七皂苷Ft1对Siha细胞增殖、迁移和侵袭的影响。流式细胞术、Hoechst33342染色实验检测细胞凋亡情况,RT-qPCR检测Bax、Bcl-2、Caspase3 mRNA水平,Western blot实验检测Bax、Bcl-2、Caspase3、cleaved-Caspase3蛋白水平。结果:与对照组相比,低、中、高浓度组Siha细胞增殖活性、细胞克隆率、细胞迁移率显著下降,侵袭细胞数显著减少,细胞凋亡率、Bax mRNA和蛋白表达水平显著升高,Bcl-2、Caspase3 mRNA和蛋白表达水平显著下降,cleaved-Caspase3蛋白表达水平显著升高,差异均有统计学意义(P均<0.05)。结论:三七皂苷Ft1可能通过Bax/Bcl-2/Caspase3通路发挥促进Siha细胞凋亡的作用。Objective:To study the effect and mechanism of Notoginsenoside Ft1 on migration,invasion and apoptosis of Siha cells.Methods:Siha cells were divided into control,low-,medium-,and high-dose notoginsenoside Ft1(0,20,40,80μmol/L),the effects of Notoginsenoside Ft1 on proliferation,migration and invasion of Siha cells were investigated by plate cloning test,cell scratch test and transwell test.Hoechst 33342 staining and flow cytometry assays were used to detect the apoptosis rate of Siha cells.The mRNA expression of Bax,Bcl-2 and Caspase3 was detected by RT-qPCR,and the protein expression of Bax,Bcl-2,Caspase3 and cleaved-Caspase3 was detected by Western blot experiment.Results:Compared with the control group,the proliferation activity,cell clone rate and cell migration rate of Siha cells in low-,medium-and high-dose groups were significantly decreased,and the number of invasive cells was significantly decreased,the apoptosis rate and the expression levels of Bax mRNA and protein were significantly increased,the levels of Bcl-2 and Caspase3 mRNAs and proteins were significantly decreased,and the levels of cleaved-Caspase3/Caspase3 protein was significantly increased(P<0.05).Conclusion:Notoginsenoside Ft1 might promote apoptosis of Siha cells through Bax/Bcl-2/Caspase3 signaling pathway.
