摘  要:目的:探究CCT2在卵巢癌中的表达及对卵巢癌细胞生物学功能的影响。方法:通过TCGA和GTEx数据库分析在卵巢癌组织及正常卵巢组织样本中CCT2的表达差异;通过ICGC数据库分析并评价CCT2表达水平与患者预后的关系。免疫组化实验验证CCT2在卵巢癌组织及癌旁正常组织的表达差异。使用Western Blot实验对比正常卵巢细胞和卵巢癌细胞系CCT2表达水平。通过转染siRNA和质粒的方式构建CCT2沉默和CCT2过表达模型,并使用qPCR和Western Blot验证。利用CCK8及平板克隆方法检测CCT2对卵巢癌细胞增殖能力的影响,同时应用Western Blot验证细胞增殖相关蛋白(Ki67、PCNA)以及EMT相关蛋白(E-cadherin、N-cadherin、Vitmentin)表达水平。使用Transwell法明确CCT2对卵巢癌细胞转移和侵袭能力的影响。并进一步通过裸鼠体内成瘤实验观察CCT2对肿瘤生长的影响。结果:CCT2在卵巢癌组织中高表达且与患者预后负相关。沉默CCT2可显著抑制卵巢癌细胞增殖、迁移和侵袭能力,反之亦然。体内成瘤实验结果证实CCT2沉默可有效减慢肿瘤生长速度。结论:CCT2在卵巢癌组织中高表达,可作为患者预后不良的分子标志物,可通过促进细胞增殖转移促进卵巢癌的进展。Objective:To investigate the expression of CCT2 in ovarian cancer and its effect on biological function of ovarian cancer cells.Methods:The expression divergence of CCT2 in ovarian cancer tissues and normal ovarian tissue samples was analyzed from TCGA and GTEx database.The relationship between the expression level of CCT2 and the patient's prognosis was evaluated using ICGC database.Immunohistochemistry was performed to detect CCT2 expression in ovarian cancer and adjacent tissues.The expression of CCT2 in normal ovarian cells and ovarian cancer cell lines was compared by Western Blot.CCT2 silencing and CCT2 overexpression models were constructed by transfecting of siRNA and plasmid and verified using qPCR and Western Blot.CCK8 and clone formation assays were performed to explore the effect of CCT2 in ovarian cancer cell proliferation.Expression of proliferation-related proteins(Ki67,PCNA)and EMT-related proteins(E-cadherin,N-cadherin,Vitmentin)was analyzed by Western Blot.Transwell assays were performed to explore the effect of CCT2 in ovarian cancer cell migration and invasion.Tumorigenicity assays in nude mice were performed to evaluate the effects of CCT2 on the tumor growth.Results:CCT2 was highly expressed in ovarian cancer tissues and the expression level of CCT2 gene was negatively correlated with the prognosis of ovarian cancer patients.Furthermore,CCT2 silencing could suppress the proliferation,migration and invasion abilities of ovarian cancer cells and vice versa.The results from in vivo experiments indicated that silencing of CCT2 inhibited tumor growth.Conclusion:The high expression level of CCT2 in ovarian cancer tissue can be used as a molecular marker for patients with poor prognosis.It can promote the progression of ovarian cancer by promoting cell proliferation and metastasis.
