摘 要:目的观察辣椒辣素对炎性疼痛大鼠的镇痛作用,探讨其镇痛作用的可能机制。方法将26只大鼠按照随机数字法分为4组,分别是对照组5例、模型组7例、低剂量组7例和高剂量组7例。模型组和给药组的大鼠在足跖中部皮下注射100μL弗氏完全佐剂(complete Freund’s adjuvant,CFA),建立慢性炎性痛模型,动物造模后第2 d行硬膜外注射辣椒辣素,观察炎性疼痛大鼠机械痛阈,用丙酮喷雾法诱发疼痛并对疼痛程度进行评分。采用免疫组化SP法检测大鼠的背根神经节(dorsal root ganglion,DRG)组织中辣椒辣素受体(transient receptor potential vanilloid-1,TRPV1)蛋白的表达情况。结果辣椒辣素提高了模型组大鼠机械痛阈和冷痛阈(P<0.05),降低了大鼠的DRG组织中TRPV1蛋白的表达。结论辣椒辣素对CFA诱导的大鼠慢性炎性痛有镇痛作用,其机制可能与下调TRPV1蛋白的表达有关。Objective e To observe the analgesic effect of capsaicin on rats with inflammatory pain,and to explore its possible mechanism.Methods Twenty-six rats were divided into 4 groups according to random number method,namely control group(5 cases),model group(7 cases),low-dose group(7 cases)and high-dose group(7 cases).The rats in the model group and the administration group were subcutaneously injected with 100μL complete Freund's ad-juvant(CFA)in the middle of the foot plantar to establish a chronic inflammatory pain model.The mechanical pain threshold of sexual pain rats was induced by acetone spray method and the pain degree was scored.The expression of transient receptor potential vanilloid-1(TRPV1)pro-tein in dorsal root ganglion(DRG)tissues of rats was detected by immunohistochemical SP method.Results Capsaicin increased the mechanical pain threshold and cold pain threshold of the rats in the model group(P<0.05),and decreased the expression of TRPV1 protein in the DRG tissue of the rats.Conclusion Capsaicin has analgesic effect on the chronic inflam⁃matory pain induced by CFA in rats,and its mechanism may be related to the down⁃regulation of TRPV1 protein expression.
