摘 要:目的诺如病毒(Norovirus,NoV)是引起全球非细菌性胃肠炎的主要病原体,严重威胁人类健康。本研究旨在分析2019—2020年西宁市区生活污水中诺如病毒GⅡ型的基因特征,了解城市污水中诺如病毒GⅡ型的流行情况。方法2019—2020年每月连续采集西宁市湟水河上游和下游水各24份,使用滤膜法富集病毒,用实时荧光定量PCR法进行病原学鉴定,检测为GⅡ型诺如病毒核酸阳性的标本进一步进行核苷酸序列分析以确定基因型,并进行同源性和系统进化分析。结果24份湟水河上游水样中未检测到诺如病毒GⅡ型,湟水河下游采集的样本中,16份阳性,阳性率为66.67%。获得14条核苷酸序列,分别属于5个基因型,包括GⅡ.P4、GⅡ.P7、GⅡ.P8、GⅡ.P12、GⅡ.P16。结论西宁市区湟水河下游水中检测到诺如病毒,并存在不同的GⅡ基因型。Objective Norovirus(NoV)stands out as the predominant agent causing global non-bacterial gastroenteritis,posing a substantial threat to human health.This study aimed to analyze the genetic characteristics of type GⅡof NoV in domestic sewage in Xining from 2019 to 2020 and understand the epidemic situation of type GⅡof NoV in the city′s sewage system.Methods From 2019 to 2020,a monthly collection of 24 water samples from both the upper and lower reaches of the Huangshui River in Xining was undertaken.The virus was enriched by the filter membrane method,and the pathogen was identified by real-time fluorescence quantitative PCR.The samples that tested positive for type GⅡ of NoV nucleic acid were further sequenced to determine the genotype,and the homology and phylogenetic analysis were performed.Results No type GⅡ of Norovirus was detected in the 24 water samples collected from the upper reaches of the Huangshui River.In contrast,16 samples collected from the lower reaches of the same river tested positive,with a positive rate of 66.67%.Fourteen nucleotide sequences were obtained,which belonged to 5 genotypes,including GⅡ.P4,GⅡ.P7,GⅡ.P8,GⅡ.P12,GⅡ.P16.Conclusion Norovirus is identified in the lower reaches of the Huangshui River in Xining,with the presence of diverse GⅡ genotypes.
