低频超声联合微泡介导曲安奈德靶向释放在大鼠外伤性视神经病变中的应用研究

摘  要:目的探讨低频超声联合微泡(LUSMB)介导曲安奈德(TA)靶向释放在大鼠外伤性视神经病变中对视网膜神经节细胞(RGCs)的保护和再生作用。方法选取20只SD大鼠,建立视神经钳夹损伤模型,随机均分为4组,分别标记为单纯损伤组(Control组)、药物组(TA组)、超声微泡组(MU组)、超声联合TA微泡组(TAMU组),大鼠以右眼为实验眼,左眼为空白对照组(NC组)。药物及微泡混悬后通过Tenon’s囊注射,术后第1天对MU组及TAMU组实施低频超声处理。术后第14天,利用视网膜免疫荧光铺片染色计数并统计存活率,苏木精-伊红染色观察视神经病理改变,CTB标记评估视神经轴突再生情况。结果视网膜免疫荧光铺片显示与Control组对比,所有治疗组RGCs存活率更高(P<0.01);其中TAMU组的存活率较TA组、MU组更高(P<0.05)。HE染色病理提示TAMU组能相对减轻病理改变,减少炎症细胞向损伤部位的浸润。但CTB顺行标记显示治疗组与Control组之间再生轴突数量无明显统计学意义(P>0.05)。结论LUSMB能促进TA在眼后段组织的靶向释放,减轻视神经损伤的炎症反应,延缓RGCs凋亡。Objective To explore the protective and regenerative effect of low-intensity ultrasound with microbubbles(LUSMB)combined with triamcinolone acetonide(TA)targeted release for retinal ganglion cells(RGCs)in rats traumatic optic neuropathy models.Methods A total of 20 Sprague-Dawley(SD)rats were randomly divided into the following 4 groups,including injury only group(Control group),drug group(TA group),ultrasound microbubbles group(MU group),ultrasound microbubbles combined with TA group(TAMU group).In each group,the right eye was used as the experimental eye,and the left eye was used as the blank control group.The drugs were mixed with microbubbles,which were injected through Sub-Tenon's Capsule.The MU group and TAMU group were treated by low-intensity ultrasound on the first day after the operation.On the 14th day after the operation,the optic nerve and retina of the rats were taken out.The quantity and survival rate of RGCs were measured by retinal immunofluorescence staining.The hematoxylin-eosin(HE)staining was used to observe the pathological changes of optic nerve and the quantity of axonal regeneration was evaluated using CTB tracking of optic nerve axons.Results The results of retinal immunofluorescence showed that optic nerve injury resulted in the least percentage of surviving RGCs in Control group for all injured group(P<0.01).Compared with TA group and MU group,TAMU group displayed significantly higher surviving rate of RGCs(P<0.05).The result of HE staining showed that TAMU groups alleviated the pathological changes and alleviated infiltration of inflmammatory cells.The results of CTB tracking indicated that the number of regenerated axons of Control group was not statistically significant compared with those of the treatment groups(P>0.05).Conclusions Low-intensity ultrasound with microbubbles can promote the infiltration and absorption of TA in the posterior ocular tissue,reduce the inflammatory response of the optic nerve injury and delay the apoptosis of RGCs.
