摘 要:以金花葵花为原料,采用超声辅助提取法提取了金花葵花黄酮,通过单因素实验和正交实验优化了提取工艺,评估了最佳工艺条件下提取的金花葵花黄酮的抗氧化活性及安全性。结果表明,金花葵花黄酮最佳提取工艺为:料液比(g∶m L)1∶15、体积分数80%的乙醇水溶液为溶剂、超声功率300 W、超声时间35 min、提取温度75℃,在该条件下,金花葵花黄酮提取率为89.97%。质量浓度为1.00 g/L金花葵花黄酮对1,1-二苯基-2-苦基苯肼自由基、羟基自由基和2,2’-联氮-双(3-乙基-苯并噻唑-6-磺酸)自由基的清除率最高,分别为94.81%、95.24%、82.80%;对三者的半数抑制质量浓度(IC_(50))分别为106.00、70.06和213.80μg/m L。金花葵花黄酮质量浓度≤0.4 g/L时,其对红细胞溶血率均<10%,属于轻度刺激;质量浓度为1.00 g/L的金花葵花黄酮对鸡胚绒毛尿囊膜无溶血。Flavonoids from Hibiseu manihot L.flower were extracted by ultrasonic-assisted extraction method,which was optimized by single factor and orthogonal experiment.The Hibiseu manihot L.flower extracted under the optimal process conditions were then evaluated for its antioxidant activity and safety.The results showed that the optimum extraction process conditions of flavonoids from Hibiseu manihot L.flower were as follows:ultrasonic power 300 W,ethanol volume fraction in ethanol-water 80%,ultrasonic time 35 min,extraction temperature 75℃,solid-liquid ratio(g∶mL)1∶15.Under the above-mentioned optimum extraction process conditions,the maximum extraction rate of flavonoids from Hibiseu manihot L.flower was 89.97%.Flavonoids from Hibiseu manihot L.flower with a mass concentration of 1.00 g/L exhibited the highest scavenging rates on 1,1-diphenyl-2-picrylhydrazyl radical,hydroxyl radical and diammonium 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfpnic acid)free radical,which were 94.81%,95.24%and 82.80%,respectively,with the corresponding half maximal inhibitory mass concentration(IC_(50))of 106.00,70.06 and 213.80μg/mL,respectively.Flavonoids from Hibiseu manihot L.flower with a mass concentration of less than 0.4 g/L showed a hemolysis rate of less than 10%on red blood cells,indicating mild irritation,while flavonoids from Hibiseu manihot L.flower with a mass concentration of 1.00 g/L displayed no hemolysis on chick embryo chorioallantoic membrane.
