摘  要:目的探讨槟榔碱对口腔黏膜下纤维化(OSF)核因子-κB(NF-κB)、转化生长因子-β_(1)(TGF-β_(1))信号通路的影响。方法选取10份上颚正常口腔黏膜样本,随机分为观察组和对照组,各5份。均经细胞培养后,观察组第6代口腔黏膜成纤维细胞(OMFb)内加入40μg/ml浓度的槟榔碱溶液,对照组加入含10%胎牛血清的DMEM培养液。比较两组样本中NF-κB、TGF-β_(1)mRNA表达、蛋白含量及12、24、48 h后细胞增殖情况。结果观察组样本NF-κB、TGF-β_(1)mRNA表达分别为(2.04±0.31)、(1.54±0.22),显著高于对照组的(1.47±0.23)、(1.17±0.18),差异具有统计学意义(P<0.05)。观察组样本NF-κB、TGF-β_(1)蛋白含量分别为(123.56±3.56)、(90.14±4.15)kDa,均显著高于对照组的(77.45±2.14)、(54.11±2.18)kDa,差异具有统计学意义(P<0.05)。观察组样本24、48 h后细胞增殖数量分别为(320.82±48.59)×10^(9)、(380.45±37.85)×10^(9),显著低于对照组的(600.76±45.37)×10^(9)、(1200.98±100.95)×10^(9),差异具有统计学意义(P<0.05)。结论高浓度槟榔碱可能通过上调NF-κB及TGF-β_(1)通路促进口腔黏膜下纤维性变,对后续的临床研究具有重要意义。Objective To explore the influence of arecoline on Nuclear factor-κB/Transforming growth factor-β_(1)(NF-κB/TGF-β_(1))signaling pathways in oral submucous fibrosis(OSF).Methods 10 samples of normal oral mucosa of the upper jaw were randomly divide into an observation group and a control group,each with 5 samples.After cell culture,the 6th generation oral mucosal fibroblasts(OMFb)of the observation group were supplemented with arecoline solution at a concentration of 40μg/ml,and the control group was supplemented with DMEM culture solution containing 10%fetal bovine serum.NF-κB,TGF-β_(1) mRNA expression,protein content and cell proliferation after 12,24 and 48 h were compared between the two groups.Results The mRNA expressions of NF-κB and TGF-β_(1) in the observation group were(2.04±0.31)and(1.54±0.22),which were significantly higher than(1.47±0.23)and(1.17±0.18)in the control group,and the difference was statistically significant(P<0.05).The protein contents of NF-κB and TGF-β_(1) in the observation group were(123.56±3.56)and(90.14±4.15)kDa,which were significantly higher than(77.45±2.14)and(54.11±2.18)kDa in the control group,and the difference was statistically significant(P<0.05).The number of cell proliferation in the observation group were(320.82±48.59)×10^(9)and(380.45±37.85)×10^(9)after 24 and 48 h,which were significantly lower than(600.76±45.37)×10^(9)and(1200.98±100.95)×10^(9)in the control group,and the difference was statistically significant(P<0.05).Conclusion High concentrations of arecoline may promote oral submucosal fibrosis through up-regulation of NF-κB and TGF-β_(1) pathways,which is of great significance for subsequent clinical studies.
