摘  要:目的:观察肝郁对不同月龄雌性大鼠学习记忆能力及海马突触可塑性的影响。方法:将不同月龄雌性大鼠,随机分为青年对照组、青年肝郁组、中年对照组及中年肝郁组。其中,青年肝郁组和中年肝郁组大鼠接受为期8周的慢性不可预知温和刺激;通过糖水偏好实验、旷场实验评价肝郁证模型;通过水迷宫和新物体识别实验评价大鼠学习记忆;尼氏染色观察海马神经元损伤情况;高尔基染色法观察海马区神经元树突损伤情况;实时荧光定量PCR法检测各组大鼠海马PSD95、MecP2、P250GAP、GAP43 mRNA表达情况。结果:相较同月龄对照组,肝郁组大鼠糖水偏好率、旷场水平运动总距离、中央区域停留时间、空间探索实验平台所在象限活动路程和时间、新物体探索次数、新物体识别指数、PSD95、MecP2、P250GAP、GAP43 mRNA表达均显著降低(P<0.01,P<0.05)。结论:肝失疏泄可能是通过下调突触可塑性相关基因,损伤海马神经元,进而影响雌性大鼠的学习和记忆能力。Objective:To observe the effects of liver depression on learning and memory ability and hippocampal synaptic plasticity in female rats of different ages.Methods:Female rats of different ages were randomly divided into a young control group,a young liver depression group,a middle-aged control group,and a middle-aged liver depression group.Among them,young and middle-aged rats with liver depression received chronic unpredictable mild stimulation for 8 weeks;Evaluate the liver depression syndrome model through sugar water preference experiments and open field experiments;Evaluate rat learning and memory through water maze and new object recognition experiments;Nissl staining was used to observe the damage of hippocampal neurons;Observation of dendritic damage in hippocampal neurons using Golgi staining method;Real time fluorescence quantitative PCR was used to detect the mRNA expression of PSD95,MecP2,P250GAP,and GAP43 in the hippocampus of rats in each group.Results:Compared with the same month old control group,the glucose preference rate,total distance of open field horizontal movement,central region residence time,quadrant activity distance and time of spatial exploration experimental platform,number of new object explorations,new object recognition index,PSD95,MecP2,P250GAP,GAP43 mRNA expression in the liver depression group rats were significantly reduced(P<0.01,P<0.05).Conclusion:Liver dysfunction and laxity may be caused by downregulating synaptic plasticity related genes,damaging hippocampal neurons,and thereby affecting the learning and memory abilities of female rats.
