摘 要:目的:探讨食管鳞状细胞癌(Esophageal Squamous Cell Carcinoma,ESCC)组织中类病毒m6A相关甲基转移酶(Vir Like M6A Methyltransferase Associated,VIRMA)的蛋白表达与患者临床病理资料的关系及其启动子区的甲基化改变。方法:免疫组织化学染色法检测40例ESCC组织和对应癌旁正常组织中VIRMA的表达,分析VIRMA的表达与ESCC患者临床病理资料的关系;运用基质辅助激光解析电离时间飞行质谱仪(MALDI-TOF MS)检测10例ESCC组织和8例癌旁正常组织中VIRMA基因启动子区的甲基化状态,初步探讨相关机制。结果:VIRMA蛋白的表达主要位于ESCC的细胞核,其在ESCC中的表达明显高于癌旁正常组织(P<0.001);VIRMA高表达在相对年轻(<60岁)及有淋巴结转移的患者ESCC组织中占比高,差异具有统计学意义(P<0.05);VIRMA蛋白的表达在患者不同性别、肿瘤发生部位、肿瘤分级、TNM分期间差异不具有统计学意义(P均>0.05);ESCC组织中VIRMA基因启动子区整体甲基化水平(0.074±0.023)低于癌旁正常组织(0.080±0.014),但差异无统计学意义(P>0.05);ESCC组织和癌旁正常组织中VIRMA基因启动子区各个CpG单位间的甲基化差异不具有统计学意义(P均>0.05)。结论:VIRMA蛋白在ESCC组织中高表达,且与食管癌的发病年龄和淋巴结转移有关,但其基因启动子区甲基化水平未见明显变化。Objective:To investigate the relationship between the expression of Vir Like M6A Methyltransferase Associated(VIRMA)in esophageal squamous cell carcinoma(ESCC)tissue and the clinicopathological data of patients and the methylation of its promoter.Methods:Immunohistochemical staining was used to detect the expression of VIRMA in 40 ESCC tissues and the corresponding adjacent normal tissues.The relationship between the expression and clinicopathological features was analyzed in combination with clinicopathological data of ESCC patients.Matrix-assisted laser desorption ionization time-of-flight mass spectrometry(MALDI-TOF MS)was used to detect the methylation of VIRMA promoter in 10 ESCC tissues and 8 adjacent normal tissues,and preliminarily explore the relevant mechanism.Results:The expression of VIRMA was mainly located in the nucleus of ESCC,and its expression in ESCC was significantly higher than that in adjacent normal tissues(P<0.001).The high expression of VIRMA was significantly higher in ESCC tissues of relatively young(<60 years old)and patients with lymph node metastasis(P<0.05).There were no significant differences in gender,tumor location,tumor grade,and TNM stage between or among the ESCC patients(all P>0.05).The total methylation level of the VIRMA promoter was lower in ESCC(0.074±0.023)compared to the adjacent normal tissues(0.080±0.014),but there was no significant difference between them(P>0.05).The methylation level of the VIRMA promoter between the individual CpG units in ESCC and adjacent normal tissues have no significant difference(all P>0.05).Conclusion:VIRMA is highly expressed in ESCC tissues and is related to the age and lymph node metastasis of esophageal cancer.However,there is no significant alteration in the methylation level of its promoter region.
