摘  要:目的探讨七氟烷预处理对1-甲基-4苯基-1,2,3,4-四氢吡啶(MPTP)诱导的急性帕金森病(PD)小鼠的神经保护作用。方法选取36只SPF级雄性C57BL/6小鼠并随机分为4组,每组9只。七氟烷组暴露于2%七氟烷2h,PD组腹腔注射MPTP20mg/kg,七氟烷预处理+PD组预先暴露于2%七氟烷2h,复苏后于小鼠腹腔注射MPTP,对照组腹腔注射等剂量0.9%氯化钠溶液。比较4组行为学指标、黑质区酪氨酸羟化酶(TH)阳性神经元数量、脑组织炎症因子[白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)]水平、氧化应激指标[丙二醛(MDA)、谷胱甘肽(GSH)]水平。结果PD组、七氟烷预处理+PD组悬挂试验掉落潜伏期均短于对照组、七氟烷组,悬挂试验评分低于对照组、七氟烷组,爬杆试验转向时间、总时间均长于对照组、七氟烷组,且七氟烷预处理+PD组悬挂试验掉落潜伏期长于PD组,悬挂试验评分高于PD组,爬杆试验转向时间、总时间均短于PD组,差异有统计学意义(P<0.05)。PD组、七氟烷预处理+PD组黑质区TH阳性神经元数量均少于对照组、七氟烷组,七氟烷预处理+PD组黑质区TH阳性神经元数量多于PD组,差异有统计学意义(P<0.05)。PD组、七氟烷预处理+PD组脑组织IL-1β、IL-6、TNF-α水平均高于对照组、七氟烷组,七氟烷预处理+PD组脑组织IL-1β、IL-6、TNF-α水平低于PD组,差异有统计学意义(P<0.05)。PD组、七氟烷预处理+PD组脑组织MDA水平均高于对照组、七氟烷组,GSH水平低于对照组、七氟烷组,且七氟烷预处理+PD组脑组织MDA水平低于PD组,GSH水平高于PD组,差异有统计学意义(P<0.05)。结论七氟烷预处理对MPTP诱导的急性PD模型小鼠具有保护作用,可改善运动功能,其作用机制可能与七氟烷预处理的抗炎、抗氧化应激相关。Objective To investigate the neuroprotective effect of sevoflurane preconditioning on 1-methyl-4-phenyl-1,2,3,6-tetrahydropyri-dine(MPTP)induced acute Parkinson's disease(PD)in mice.Methods 36 SPF male C57BL/6 mice were selected and randomly divided into 4 groups,with 9 mice in each group.The sevoflurane group was exposed to 2%sevoflurane for 2 h,the PD group was intraperitoneally injected with MPTP 20 mg/kg,the sevoflurane pretreatment+PD group was pre-exposed to 2%sevoflurane for 2 h,and the mice were intraperitoneally injected with MPTP after resuscitation,the control group was intraperitoneally injected with the equal dose of 0.9%sodium chloride solution.The behavioral indexes,the number of tyrosine hydroxylase(TH)positive neurons in substantia nigra,the levels of inflammatory factors(interleukin-1β[IL-1β],in-terleukin-6[IL-6],tumor necrosis factor-α[TNF-α])and oxidative stress(malondialdehyde[MDA]and glutathione[GSH])in brain tissues were com-pared among the four groups.Results The falling latency of suspension test in the PD group and the sevoflurane pretreatment+PD group was short-er than that in the control group and the sevoflurane group,the suspension test score was lower than that in the control group and the sevoflurane group,the turning time and total time of pole climbing test were longer than those in the control group and the sevoflurane group,and the falling la-tency of suspension test in the sevoflurane pretreatment+PD group was longer than that in the PD group,the suspension test score was higher than that in the PD group,the turning time and total time of pole climbing test were shorter than those in the PD group,the differences were statistically significant(P<0.05).The number of TH positive neurons in substantia nigra in the PD group and the sevoflurane preconditioning+PD group was less than that in the control group and sevoflurane group,and the number of TH positive neurons in substantia nigra in the sevoflurane pretreatment+PD group was more than that in the PD group,the differences w
