摘 要:目的通过建立脊髓栓系综合征(tethered cord syndrome,TCS)大鼠模型,观察后肢胫后肌群骨骼肌萎缩的早期病理改变,探讨miRNA-206在此过程中的变化及其对骨骼肌萎缩的影响。方法采用手术方法制备TCS大鼠模型。观察造模与拮抗剂处理后大鼠后肢胫后肌群骨骼肌萎缩的早期功能障碍和肌肉体积、肌湿重(比)、肌纤维平均横截面积的变化,并采用实时荧光定量聚合酶链反应检测miRNA-206在大鼠后肢胫后肌群中的表达。结果术后7 d起,模型组大鼠后肢开始出现功能障碍。28 d时模型组miRNA-206表达明显升高,是假手术组的5.54倍,肌湿重、肌纤维平均横截面积减小,与假手术28 d组比较,差异有统计学意义(均P<0.05)。给予miRNA-206拮抗剂后,肌纤维平均横截面积及胫后肌群体积明显缩小,肌湿重比减少,差异有统计学意义(P<0.05)。结论TCS模型大鼠后肢胫后肌群骨骼肌萎缩的早期病理改变会造成miRNA-206表达显著增加,这可能是机体的一种自我保护机制。ObjectiveTo observe early pathological changes of skeletal muscle atrophy in posterior tibial muscle group of hind limbs by establishing a rat model of tethered cord syndrome(TCS),to explore the changes of miRNA-206 in this process and to examine its effect on skeletal muscle atrophy.MethodsThe TCS rat model was prepared surgically.The study observed early functional impairment and changes in muscle volume,wet muscle weight(ratio),as well as average cross-sectional area of muscle fibers in posterior tibial muscle group of hind limbs after modeling and antagonist dosing.Real-time fluorescent quantitative polymerase chain reaction was employed for detecting the expression of miRNA-206 in hind limbs.ResultsAs of Day 7 postoperatively,functional impairment appeared in hind limbs of model group.At Day 28,the expression of miRNA-206 spiked markedly in model group.It was 5.54 folds that of sham operation group.Wet weight and average cross-sectional area of muscle fiber declined obviously.As compared with sham operation Day 28 group,the difference was statistically significant(all P<0.05).After dosing of miRNA-206 antagonist,average cross-sectional area of muscle fiber,volume of posterior tibial muscle group and muscle wet weight ratio declined greatly.And the differences were statistically significant(P<0.05).ConclusionIn skeletal muscle atrophy of posterior tibial muscle groups in hind limbs of TCS rats,early pathological changes lead to a marked up-regulation of miRNA-206.This may represent a form of self-protection mechanism by the organism.
