摘  要:文章为研究肝损伤对大鼠体内硫胺素及其代谢物水平的影响,通过14天胆管结扎和硫胺素限制饮食分别构建肝损伤和硫胺素缺乏大鼠模型,造模成功后,测定血浆和脑皮层中硫胺素及其代谢物浓度,并进一步分析了脑皮层中焦磷酸硫胺素依赖酶的活性和蛋白表达水平。结果表明:14天胆管结扎诱导的肝损伤大鼠与硫胺素缺乏大鼠的血浆和脑皮层中硫胺素浓度显著降低,但其代谢物焦磷酸硫胺素的浓度未发生改变,脑皮层中焦磷酸硫胺素依赖酶活性和蛋白表达也未发生改变。14天胆管结扎诱导的肝损伤和硫胺素限制饮食均会显著降低大鼠血浆和脑硫胺素水平,但不影响其代谢物焦磷酸硫胺素浓度及其依赖酶的活性和表达。To investigate the effects of liver injury on the levels of thiamine and its metabolites in rats,14-day bile duct ligation and thiamine-restricted diet were used to construct liver injury and thiamine-deficient rat models,respectively.After successful modeling,the concentrations of thiamine and its metabolites in plasma and cerebral cortex were measured,and the activity of thiamine pyrophosphate-dependent enzymes and protein expression levels in cerebral cortex were further analyzed.14-day bile duct ligation induced liver injury in rats and thiamine-deficient rats resulted in a significant decrease in the concentration of thiamine in the plasma and cerebral cortex,but the concentration of its metabolite,thiamine pyrophosphate,remained unchanged.The activity of thiamine pyrophosphate-dependent enzymes and protein expression in the cerebral cortex also did not change.14-day bile duct ligation induced liver injury and thiamine-restricted diet significantly reduced plasma and brain thiamine levels in rats,but did not affect the concentration of its metabolite thiamine pyrophosphate as well as its dependent enzyme activity and expression.
