摘  要:目的 构建红景天苷脂质体(SAL-LIPS),对其进行评价,并研究SAL-LIPS在高糖(HG)环境下对人视网膜色素上皮(ARPE-19)细胞氧化损伤的保护作用。方法 采用乙醇注入法制备SAL-LIPS,通过透射电镜观察其形态,测定粒径、多分散系数(PDI)并考察制剂的体外释放及稳定性。HG诱导ARPE-19细胞建立氧化应激细胞模型,检测不同浓度的SAL-LIPS对损伤细胞活力的影响,并检测不同组别细胞ROS水平、MDA含量、血管内皮因子(VEGF)和缺血诱导因子(HIF-1α)的表达。结果 制备的SAL-LIPS呈圆球状,平均粒径为(112.1±2)nm,平均PDI为0.204±0.02,与红景天苷原料药相比具有缓释作用,稳定性较好。在50~100 μmol·L^(-1)内SAL-LIPS可以显著改善HG诱导的ARPE-19细胞的活力。与空白对照组比较,HG组显著升高ARPE-19细胞内ROS水平、MDA含量,同时增强了VEGF和HIF-1α的表达;与HG组ARPE-19细胞比较,HG+低浓度SAL-LIPS组和HG+高浓度SAL-LIPS组显著降低了细胞内ROS水平、MDA含量,同时减少了VEGF和HIF-1α的表达。结论 所构建的SAL-LIPS具有良好的缓释作用和稳定性,可以改善HG诱导ARPE-19细胞发生的氧化应激,发挥抗氧化作用。Objective To construct salidroside liposomes(SAL-LIPS),evaluate their protective effects against oxidative damage in human retinal pigment epithelial(ARPE-19)cells under high glucose conditions.Methods SAL-LIPS were prepared with ethanol injection method.The morphology was observed with transmission electron microscopy,and particle size,polydispersity index(PDI),as well as in vitro release and stability assessed.An oxidative stress cell model was established in ARPE-19 cells induced by high glucose.The effect of different concentrations of SAL-LIPS on the viability of damaged cells was examined,and the expression levels of reactive oxygen species(ROS),malondialdehyde(MDA),vascular endothelial growth factor(VEGF),and hypoxia-inducible factor(HIF-1α)in different groups were measured.Results The prepared SAL-LIPS were spherical with an average particle size of(112.1±2)nm and an average PDI of 0.204±0.02,demonstrating a sustained-release effect and better stability compared to the raw material of salidroside.SAL-LIPS greatly improved the viability of ARPE-19 cells induced by high glucose of 50~100μmol·L^(-1).Compared with the blank control group of ARPE-19 cells,the high glucose group obviously increased the levels of intracellular ROS and MDA,along with enhanced expression of VEGF and HIF-1α.Compared with the high glucose group of ARPE-19 cells,both the low and high concentration SAL-LIPS groups much reduced the levels of intracellular ROS and MDA,and also decreased the expression of VEGF and HIF-1α.Conclusion The constructed SAL-LIPS exhibit good sustained-release effect and stability,which can improve oxidative stress induced in ARPE-19 cells by high glucose,exerting antioxidative effect.
