摘  要:目的:采用核磁共振测定三硝基苯磺酸诱导的慢性肠炎大鼠机体内源性代谢物的变化,探讨慢性肠炎的疾病机制。方法:30只SD大鼠随机分为慢性肠炎组(CC组)和空白对照组(NC组),酶联免疫吸附测定法(ELISA)检测小鼠血清中髓过氧化物酶(MPO)、IL-2和IL-4水平;核磁共振法(1H NMR)检测大鼠尿样和粪样中内源性代谢物水平。结果:与NC组比较,CC组MPO、IL-2表达均显著升高,IL-4表达显著降低,尿样核磁谱图中支链氨基酸(亮氨酸和异亮氨酸)、甘氨酸、苯丙氨基酸上调,而富马酸、牛磺酸、马尿酸、乙酰乙酸等显著下调;粪样中的甘油磷脂胆碱、丝氨酸等明显下调,尿嘧啶、肌酸等显著升高,主要涉及到氨基酸代谢、脂质代谢等代谢途径。结论:慢性肠炎发病与氨基酸、脂质代谢紊乱有关,其疾病机制可能与氨基酸代谢途径被抑制及脂质中甘油磷脂类代谢异常相关联。Objective To evaluate the changes in endogenous metabolites in rats with trinitrobenzene sulfonic acid-induced chronic colitis using nuclear magnetic resonance(NMR)and to elucidate the underlying mechanisms of chronic colitis.Methods Thirty SD rats were randomly assigned to a chronic colitis group(CC group)and a normal control group(NC group).Serum levels of myeloperoxidase(MPO),interleukin-2(IL-2),and interleukin-4(IL-4)were quantified u⁃sing enzyme-linked immunosorbent assay(ELISA).1H NMR was used to measure the endogenous metabolites in rat urine and fecal samples.Results Compared with the NC group,the CC group exhibited increased MPO and IL-2,but decreased IL-4,along with up-regulation of branched-chain amino acids(leucine and isoleucine),glycine and phenylpropyl amino acids in urine NMR,while significant down-regulation of fumaric acid,taurine,hippuric acid and acetoacetic acid.In fe⁃cal samples from the CC group,glycerophospholipid choline and serine were significantly reduced,while uracil and creatine were markedly elevated,mainly involving amino acid and lipid metabolism.Conclusion Chronic colitis is related to the dis⁃order of amino acid and lipid metabolism,and the mechanism of which may be associated with the inhibition of amino acid metabolic pathways and the abnormal metabolism of glycerophospholipids.
