摘  要:目的探究黄芩苷通过抑制IL1R2在A549肺癌细胞增殖、迁移及凋亡中的作用,并探究可能作用机制。方法体外培养A549肺癌细胞,转染si-IL1R2质粒分为对照组、低表达IL1R2组、黄芩苷+低表达IL1R2组、过表达IL1R2组、黄芩苷+过表达IL1R2组,对照组正常培养细胞,含有黄芩苷的实验组采用200mol/L的黄芩苷处理24h。实时荧光定量PCR(qRT-PCR)法测定IL1R2表达;采用CCK-8法、Transwell、流式细胞法分别观察细胞增殖、迁移能力及凋亡情况;采用Western Blotting法观察各组白介素-1β(IL-1β)、白介素-6(IL-6)、肿瘤坏死因子-ɑ(TNF-ɑ)表达。结果IL1R2低表达组干扰后A549细胞的增殖受到抑制,IL1R2过表达组则明显促进细胞增殖;低表达IL1R2组较对照组细胞迁移个数降低,过表达IL1R2组细胞迁移个数较对照组升高,与低表达IL1R2组细胞迁移个数相比,黄芩苷+低表达IL1R2组细胞迁移个数降低明显;与过表达IL1R2组迁移个数相比,黄芩苷+过表达IL1R2组细胞迁移个数降低;与对照组相比,过表达IL1R2组、黄芩苷+过表达IL1R2组细胞凋亡率均降低;黄芩苷+过表达IL1R2组较过表达IL1R2组细胞凋亡率升高;低表达IL1R2组IL-1β、IL-6、TNF-α等细胞因子表达明显下降,过表达IL1R2组各组细胞因子表达明显升高,黄芩苷+过表达IL1R2组各组细胞因子表达较过表达IL1R2组降低。结论黄芩苷通过抑制IL1R2表达抑制肺癌细胞的A549肺癌细胞增殖、迁移,促进肺癌细胞凋亡。Objective To explore the effects of baicalin on the proliferation,migration,and apoptosis of A549 lung cancer cells by inhibiting IL1R2,and to explore the possible mechanism of action.Methods A549 lung cancer cells were cultured in vitro and transfected with si-IL1R2 plasmid and divided into a control group,low-expression IL1R2 group,baicalin+low-expression IL1R2 group,over-expression IL1R2 group and baicalin+over-expression IL1R2 group.The experimental group containing baicalin was treated with 200mol/L baicalin for 24h.Real-time quantitative fluorescence PCR(qRT-PCR)was used to express IL1R2.Cell proliferation,migration,and apoptosis were observed by CCK-8,Transwell,and flow cytometry.Western blotting method was used to observe the expressions of interleukin-1β(IL-1β),interleukin-6(IL-6),and tumor necrosis factor-alpha(TNF-α)in each group.Results The proliferation of A549 cells was inhibited in the IL1R2 low-expression group and promoted in the IL1R2 overexpression group;the number of cells migrating in the IL1R2 low-expression group decreased compared with that in the control group,while the number of cells migrating in the IL1R2 overexpression group increased compared with the control group,and the number of cells migrating in the baicalin+low-expression group was significantly lower compared with the number of cells migrating in the IL1R2 low-expression group.The number of migrating cells in the baicalin+overexpression of IL1R2 group decreased compared with the number of migrating cells in the overexpression of IL1R2 group,IL-6,TNF-α,and other cytokines were significantly decreased in the low-expression IL1R2 group;cytokine expression was significantly increased in all groups of the overexpression IL1R2 group;cytokine expression was decreased in all.Conclusion Baicalin can inhibit the proliferation and migration of A549 lung cancer cells by inhibiting the expression of IL1R2,and promoting the apoptosis of lung cancer cells.
